QbD-Driven Development and Evaluation of EGCG-Loaded Transfersomal Gel for Non-Melanoma Skin Carcinoma Singh B, Nanda S, and International Journal of Drug Delivery Technology, 2025 The most common type of cancer is non-melanoma skin cancer (NMSC), necessitating the development of non-invasive and novel method of treatment. The current research study is focused on the design, development, and evaluation of a transfersomal gel loaded with epigallocatechin-3-gallate (EGCG), a bioactive compound with significant anti-cancer properties. After establishing anticancer properties of EGCG using cell lines and in silico studies, transfersomes were formulated using Box-Behnken Design (BBD). Optimization was done with three factors and three levels The optimized transfersomal gel exhibited significant physicochemical characteristics. Smaller V.S., improved Z.P. and high EE facilitated enhanced skin penetration as proved by in vitro and ex vivo studies using porcine skin. Malik International Journal of Drug Delivery Technology, 2024 Numerous chemopreventive drugs like aloin and curcumin have demonstrated potential effects in cancer treatment; however, their efficacy is limited by low bioavailability. Thus, the primary goal of this research was to formulate a unique method by using the herbal drugs synergistically and improve the bioavailability of these anticancer drugs here, using the nanoprecipitation method for two well-active constituents of known natural herbal compounds. Aloin (AL)-curcumin (CUR) were co-loaded into polymeric nanoparticles made of poly (D, L-lactide-co-glycolide) and surfactant-polyvinyl alcohol, with the experiment’s response being evaluated using Stat-Ease software. To create dual-loaded nanoparticles containing aloin and curcumin, using different ratios sand optimizing the best formulation. (60:120). For aloin curcumin, optimize the better formulation with controlled release formulation having less than 100 nm size of particle and drug entrapment was 86.09% while for (60:60) of aloin -curcumin having 99.1 nm particle size of drug and encapsulation efficiency was 72.05%.
Oxalis corniculata and Ficus religiosa mitigates rotenone-induced Parkinson’s disease in Swiss Albino mice: Mechanistic insights and therapeutic potential Praveen Kumar, Himanshu Sachdeva, Sunishtha Kalra, Balwan Singh, Ram Dayal, et al. Journal of Applied Pharmaceutical Science, 2024 Parkinson’s disease (PD) is a neurodegenerative disorder distinguished by the presence of progressive deterioration of the nervous system and is prevalent among the elderly population. The pathological hallmark of PD involves the accumulation of the protein alpha-synuclein and neuronal inflammation, which leads to the degeneration of dopaminergic (DAergic) neurons. In the present study, neuroprotective effect of combinational extract of Ficus religiosa (F. religiosa) and Oxalis corniculata (O. corniculata) L. in a mouse PD model. By giving Rotenone (ROT) on alternate days subcutaneously for 21 days, a disease model was generated. Extracts were administered individually as well as in combination orally 1 hour before ROT (1.5 mg/kg, s.c.) administration. On day 22nd, after sacrificing of animals neurochemical assays [total protein content (TPC), malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), nitrite (NO)], neurotransmitter analysis, and histology of the brain were performed. Results of the study showed a significant increase in the level of TPC, CAT, SOD, GSH, and dopamine in substantia nigra and a significant reduction of MDA, NO, and glutamate after administration of F. religiosa and O. corniculata in combination (200 + 500 mg/kg, p.o.) in a ROT induced model. This study demonstrated that a combination of F. religiosa and O. corniculata improved motor symptoms and reduced dopaminergic neuron death in mice challenged with ROT.
Pharmacokinetic Study of Aloin Nanoparticulate: Enhanced Oral Formulation Bioavailability Rishika Chauhan, Balwan Singh, MP Singh, Anuj Malik International Journal of Pharmaceutical Quality Assurance, 2024 This research aimed to prepare and assess an aloin nanoparticle that might increase its oral bioavailability. Utilizing the nanoprecipitation process, aloin Poly lactic-co-glycolic acid (PLGA) nanoparticulate was formulated and studied for the best strategy based on their characteristics and in-vitro release. The aloin nanoparticle stability research was evaluated at room temperature for a duration of three months. Additionally, pharmacokinetic (PK) tests were analyzed by using various parameters and experiments done by taking female wistar albino rats, with weights of approximately (150–200 gm). The animal was appropriately given the proper housing according to the protocol and placed in a period of twelve hours day and night. The drug was administered for 72 hours at a dosage of 10 mg/kg. of all the aloin nanoformulations developed, the one with the best size of particles was 98.5 nm with an entanglement efficacy of 98.09%. The results show that when compared to pure aloin, aloin PLGA nanoparticulate has a higher bioavailability.
Formulation and Evaluation of Self-Emulsifying Drug Delivery Systems for Candesartan Cilexetil Balwan Singh, Manish Sharma International Journal of Pharmaceutical Sciences and Nanotechnology, 2022 Introduction: Candesartan cilexetil is an angiotensin receptor blocker prescribed for hypertension management. However the drug belonging to BCS class II has low solubility and in turn low bioavailability. Lipid-based drug delivery systems are gaining wide attention in the field of pharmaceutical formulations owing to their potential to enhance the solubility of poorly aqueous soluble drugs.
 Objective: “Present” work aimed to formulate and evaluate Candesartan cilexetil loaded Self-emulsifying drug delivery systems (SEDDS) as a potential antihypertensive drug delivery system by improving its solubility
 Methods: Formulation of drug incorporated SEDDS was carried out using various oils, surfactants, and cosurfactants. Preliminary solubility studies in these excipients were performed followed by the construction of a Pseudoternary phase diagram for optimization of all three excipient concentrations. After this, SEDDS of Candesartan were formulated and evaluated for clarity, phase separation, drug content, % transmittance, globule size, freeze-thaw, in vitro dissolution studies, and particle size analyses.
 Results: Observation from preliminary solubility studies resulted in the selection of Acrosyl k-160 (oil phase), Labrafac PG (surfactant), and Transcutol-P (co-surfactant). A pseudo ternary phase diagram was constructed to optimize the concentration ranges of chosen oil, surfactant, and co-surfactant. In total twelve formulations were prepared and evaluated for various parameters. FTIR analysis indicated negligible drug excipient interaction. CF11 was identified as the optimal formulation based on the particle size of an average of about 50.2nm, drug content (98.66%), and in vitro release profile, with a drug release of 99.41±5.79 % after one hour. The formulations were also put through thirty-day thermodynamic stability studies and were found to be stable
 Conclusion: SEDDS can be formulated to improve the dissolution and oral bioavailability of the poorly water-soluble drug Candesartan, according to the findings of this study.
