Biocontrol and growth-promoting potential of Streptomyces hygroscopicus against bacterial leaf blight in rice Titik Nur Aeny, Radix Suharjo, Sudi Pramono, Suskandini Ratih Dirmawati, Hamim Sudarsono, et al. Australian Journal of Crop Science, 2026 This study evaluated the antagonistic activity and plant growth-promoting potential of Streptomyces hygroscopicus against Xanthomonas oryzae pv. oryzae, the causal agent of bacterial leaf blight (BLB) in rice, under in vitro and greenhouse conditions. Experiments were arranged in a completely randomized design. In vitro assays compared sterile water, chloramphenicol, and S. hygroscopicus, while greenhouse trials assessed three application doses of S. hygroscopicus (5, 10, and 15 mL per plant), a bactericide, and a control treatment, each with four replications. The in vitro assay demonstrated a strong antagonistic effect of S. hygroscopicus, indicated by the formation of distinct inhibition zones against X. oryzae. Under greenhouse conditions, foliar application of S. hygroscopicus (5-15 mL per plant; 10⁸ cfu/mL) delayed disease incubation, significantly reduced disease severity and the area under the disease progress curve (AUDPC). Moreover, higher application doses (10 – 15 mL per plant) enhanced plant height and leaf number. A clear dose-dependent reduction in AUDPC was observed, indicating sustained suppression of disease development over time. Overall, the results highlight the potential of S. hygroscopicus as an effective and environmentally friendly biocontrol agent with additional plant growth-promoting benefits. Further studies under field conditions are recommended to confirm its consistency and applicability in integrated management of bacterial leaf blight in rice.

Members of the genus Streptomyces represent taxonomically diverse and metabolically versatile actinobacteria commonly associated with rhizosphere ecosystems. This study presents a polyphasic characterization of a Streptomyces isolate obtained from the pineapple (Ananas comosus) rhizosphere in Lampung, Indonesia, integrating morphological, molecular, and metabolite profiling approaches. Morphological examination using scanning electron microscopy revealed extensively branched vegetative hyphae and rectiflexibiles-type spore chains with smooth surfaces, consistent with diagnostic features of the genus. Molecular identification based on 16S rRNA gene sequencing (~1,450 bp) placed the isolate within the Streptomyces hygroscopicus clade (GenBank accession no. PZ049818), showing low genetic divergence (<0.02 substitutions per site) relative to closely related strains, although species-level resolution remains limited using 16S rRNA alone. Gas Chromatography-Mass Spectrometry (GC-MS) analysis of ethyl acetate extracts detected 53 volatile and semi-volatile compounds, with similarity indices ranging from 66% to 96%; compounds with similarity indices ≥80% were considered reliable identifications. Major constituents included 3-deoxy-D-mannoic lactone (6.92%), (S)-(+)-2-amino-3-methyl-1-butanol (6.42%), 2-methylbutanoic anhydride (5.74%), catechol (5.16%), and 9,12-octadecadienoic acid (Z,Z) (4.58%), representing diverse chemical classes such as organic acids, alcohols, phenolics, and fatty acids. Although limited to a single isolate and qualitative metabolite profiling, this study provides baseline taxonomic and metabolomic data for pineapple-associated Streptomyces, serving as a foundation for future genomic, functional, and biocontrol-related studies in tropical agroecosystems.
Relief from Salt Stress by Plant Growth-Promoting Bacteria in Hydroponic Leaf Lettuce (Lactuca sativa L.) Climate change and saltwater intrusion both endanger freshwater availability as a source of drinking water and agricultural resources. Therefore, the saline and brackish water demand for irrigated farm goods should be considered a method for producing salt-resistant crop products. This study aimed to monitor Indole-3-acetic acid (IAA) and biofilm by selected bacterial isolates and its impact on the growth of lettuce ( Lactuca sativa L.) during salt stress. Several isolates are employed such as Streptomyces hygroscopicus GGF4-i18, Streptomyces sp. AB8, Micrococcus luteus , Serratia marcescens MBC1, and Streptomyces hygroscopicus subsp. Jinggangensis InaCC A497. A 1.0 percent L-tryptophan is used as a precursor of IAA. The impact of IAA and biofilm produced by bacteria was demonstrated by inoculating bacterial isolates on Lactuca sativa (lettuce) plants in the hydroponic system. The nutrient film technique (NFT) is the selected hydroponic cultivation technique. The result shows that Streptomyces genera were able to maintain higher IAA hormone production within a week. On the other hand, Serratia marcescens MBC1 and Micrococcus luteus tended to have stable IAA levels. Microbes need a definite time to synthesize a compound. The microbial life cycle consists of several phases, often requiring a shorter incubation time than in complex organisms such as plants, animals, and humans. This study aimed to determine the optimum incubation time and the best tryptophan levels of Streptomyces hygroscopicus subsp. hygroscopicus strain i18 in.Indole-3-acetic acid (IAA) hormone. IAA has synthesized on liquid ISP4 medium using L-tryptophan supplementation as a precursor. Cell density was measured every day for 4-11 days using the spectrophotometric approach to track the incubation time. The doses of L-Tryptophan were 0, 1, 2, 3, 4, 5, and 6 mg/mL, respectively. The levels of IAA produced by bacteria were measured and compared with standard IAA. Streptomyces hygroscopicus subsp. hygroscopicus strain i18 was obtained the optimum incubation time at 9th days with an IAA level of 12.30 µg/mL. Supplementing with 5 mg/mL L-tryptophan resulted in the highest IAA level of 17.90 µg/mL. This study would be worthwhile in assisting agricultural product quality improvement through field application for further research.
In vitro test for inhibition of plasmodium falciparum 3d7 parasites using streptomyces hygroscopicus subsp. hygroscopicus strain i18, isolated from a Pineapple Farm in lampung Endah Setyaningrum, Achmad Arifiyanto, Nismah Nukmal, Titik Nur Aeny, Meishy Handerlin Putri, Ulin Ni’mah Setiawati Journal of Pure and Applied Microbiology, 2021 Increasing natural ingredient awareness and utilization has created an increased demand for sources of natural medicinal ingredients, including sources of compound used to treat malaria. Streptomyces is a genus of prokaryote well recognized for its production of antibiotics and other pharmaceutically useful compound. This study aimed to assess the ability of unpurified fermentation metabolites to inhibit Plasmodium parasites. A strain of bacteria identified as Streptomyces hygroscopicus subsp. hygroscopicus strain i18 were isolated from pineapple fields in Lampung province, and was cultured and fermented on liquid synthetic Gause medium for 10 days. The supernatant was separated from the cells and extracted with ethyl acetate-methanol (1:1). Plasmodium falciparum 3D7 was used for antiplasmodial testing. Metabolites were tested qualitatively using a phytochemical approach. Saponins and triterpenoids were found to be present in the extract. Parasite inhibition as measured using probit analysis and yielded an IC50 value of 11.07 g.m/L. These findings suggest further examinations of this extract (e.g. assessment of off-target effects) are warranted.
The Diversity and the Abundance ofCorn Planthopper(Hemiptera: Delphacidae)inLampung Province R Hasibuan, Y Fitriana, S Ratih, L Wibowo, T N Aeny, FX Susilo, I G Swibawa, F R Lumbanraja Journal of Physics Conference Series, 2021 The outbreak of delphacid planthoppers has been detected across corn-growing regions in South Lampung. Survey study was conducted in three corn fields in Natar District, South Lampung Regency. In each study site, five corn plants were randomly sampled. In each sampled plant, one leaf with maximum number of planthoppers was selected for population recording. Based on the morphological identification results, there were two types of corn planthoppers attacking corn fields during sampling periods: the white bellied-planthopper, Stenocranus pacivicus Kirkaldy and Peregrinus maidis Ashmead. During sampling periods, S. pacivicus was most abundant species, while, the Peregrinus planthopper was almost undetectable. There was similar trend peak of density S. pacificus brachypters & nymph and macropters among the three corn fields. The maximum number of S. pacificus brachypters & nymph 412.38 ±23.12 individuals’ leaf−1 was recorded at 70 days after planting (DAP). While the S. pacificus macropters reached the highest population number 43.81 ± 5,76 individuals leaf−1. Extended feeding activity by planthoppers caused the chlorosis and necrosis of the leaf, reduced plant vigor, and stunting, resulting hopperburn symptoms. The results of this study confirm that the explosion of delphacid planthopper sand corn hopperburn are real and become a threat for corn production in Lampung Province.
Simultaneous detection of virus infecting tobacco plant using multiplex-PCR in Klaten, Indonesia Selvi Helina, Titik Nur Aeny, Ivayani Ivayani Archives of Phytopathology and Plant Protection, 2021 Tobacco is one of the important plantation commodities in Indonesia. The multiple virus attack in the field causes crop damage to crop failure. Therefore, detection is needed quickly and accurately as a basis for control strategies. This study aimed to determine the viruses that attack tobacco plants in Klaten, Indonesia through simultaneous detection using multiplex-PCR. The study was conducted by observation in the field then detection in the Laboratory. Based on Multiplex-PCR detection results using primers of MJ 1/MJ 2, Krusty/Homer, and CMV P1/P2 showed that tobacco plants infected with several viruses from the genus Potyvirus, Begomovirus and Cucumovirus. Based on the results of the nucleotide sequencing blast, it was found that the tobacco isolate was positively infected by the Tomato yellow leaf curl Kanchanaburi virus (TYLCV), Cucumber mosaic virus (CMV) and Potato virus (PVY).

Biodiversitas 22: 2817-2823. Streptomyces is a potential bacterial genus that has been investigated extensively as a source of natural microbial compounds. Its potential metabolites have been widely developed for pharmaceutical, pathogen control, and other applications in agriculture. This study aimed to determine the ability of the Streptomyces sp AB8 crude extract in inhibiting Plasmodium and pathogenic microbes. Streptomyces was cultured on Gause synthetic media for 10 days. The fermented broth culture media has dissolved in a 1:1 mixture of ethyl acetate and methanol. Biochemical characterization of this isolate has carried out using the standard methods. In-vitro antimalarial activity assay was performed using a chloroquine-sensitive Plasmodium falciparum strain 3D7. Fresh type O-positive human erythrocytes were suspended at 4 percent hematocrit in a complete medium to maintain culture. The inhibitory concentration (IC50) was determined using probit analysis. The results showed the extract of Streptomyces sp. AB8 contains phenolic and alkaloids. Streptomyces sp. AB8 extract can inhibit Dickeya zeae N-Unila 5, Dickeya zeae N-Unila 10, Aspergillus sp IK3, and Escherichia coli growth. The results also showed that the ICs0 value of extract against P. falciparum 3D7 was 17.56 ug/mL. Further research was needed to determine the types of purified bioactive compounds and their bioactivity.
Erwinia mallotivora is the causal agent of papaya bacterial crown rot disease in lampung timur, indonesia Radix Suharjo, Hani Anggrainy Oktaviana, Titik Nur Aeny, Cipta Ginting, Rachmansyah Arianto Wardhana, Ariyo Nugroho, Ratdiana Ratdiana Plant Protection Science, 2021 Sixteen bacterial strains showing oval, convex with a white colony colour were obtained from the water-soaked lesions on the petioles and leaves of infected papaya (cv. calina) collected from a papaya field in Lampung Timur, Indonesia. The pathogenicity test showed that all the strains produced the same symptoms with those found in the field. Four representative strains were then chosen for further investigation. The phenotypic characteristics revealed that the strains resembled Erwinia mallotivora. Two representative strains were further identified using a 16SrDNA sequence analysis. The result showed that the strains were placed within the group of the type strain and the reference strains of E. mallotivora. To the best of our knowledge, this is the first finding of E. mallotivora as the causal agent of papaya crown rot disease in Indonesia. Among the sixteen plants used for the host range test, the symptom was only observed on eggplants, but not on the other fifteen plant species.
THE POTENCY OF CALABUR TREE (Muntingia calabura) LEAF EXTRACT TO CONTROL ANTHRACNOSE OF PAPAYA FRUIT Efri Efri, Titik Nur Aeny, Ivayani Ivayani, Meisroyatul Hulfa Journal of Tropical Plant Pests and Diseases, 2020 The potency of calabur tree (Muntingia calabura) leaf extract to control anthracnose of papaya fruit. The papaya anthracnose caused by Colletotrichum gloeosporioides is one of the factors causing a decrease in papaya production. The research aimed to determine the abilty of calabur tree leaf extract in inhibiting growth, sporulation, and viability of C. gloeosporioides as well as incubation period and disease severity on the fruit of papaya. The experiment was arranged in a nested design, the concentrations (0, 10, 20, 30, 40, 50, and 60%) were nested within the calabur tree leaf extract methods (boiling and fractionation). The results showed that boiled calabur leaves extract was more effective than fractionated calabur leaves extract to inhibit growth of C. gloeosporioides. Boiled calabur leaves extract and fractionated leaves extract at various concentration showed capability to inhibit the growth, sporulation and viability of C. gloeosporioides. Boiled calabur leaves extract at different concentration levels were able to suppress disease development of papaya anthracnose disease on papaya fruit. The higher concentration of calabur leaves extract, are more effective to inhibit C. gloeosporioides.
