Design, synthesis and biological evaluation of novel chalcone-derived thioxopyridine and pyrazolopyridine compounds as antimicrobial agents Gaber Algaber, P. Shyamala, Zaccaria Dammag, Mohammed Al-haddad Scientific Reports, 2026 The present work deals with the synthesis of some new chalcone, 2- thioxopyridine,2-methylthiopyridine, 2-hydrazinopyridine and 3-amino-1H-pyrazolo[3,4-b]pyridine derivatives containing biologically active substituents. The starting material of the present study was synthesis of the 2-cyanoethanethioamide from malononitrile using a modified Brunskill procedure, followed by the synthesis of the 1,3-diarylprop-2-en-1-one (chalcone) from a ketone and an aldehyde. 2-Thioxopyridines were obtained through Michael addition of α,β-unsaturated ketones with 2-cyanothioacetamide to afford the non-isolable intermediates 8 & 9 followed by cyclization and subsequent dehydration and dehydrogenation of the cyclized product to give compound 4-(furan-2-yl)-6-(4- methoxyphenyl)-2-thioxo-1,2-dihydropyridine-3-carbonitrile which was reacted with methyl iodide in methanolic sodium methoxide to afford the corresponding 4-(furan-2-yl)-6-(4-methoxyphenyl)-2-(methylthio) pyridine-3-carbonitrile, which was reacted with hydrazine hydrate to give the corresponding 2-hydrazinopyridines, which cyclized to afford the corresponding 3-aminopyrazolo[3,4-b]pyridine derivatives and were characterized by FT-IR, 1H-NMR, 13C-NMR and MS spectral studies. All the newly synthesized compounds were evaluated for their in vitro antibacterial activity against S. aureus as example of Gram-positive bacteria and E. coli as example of Gram-negative bacteria. They were also evaluated for their in vitro antifungal potential effect against C. albicans. The zone of inhibition was measured using the well diffusion assay, and the results indicated that compounds 3, 5, 10 and 13 showed potential antibacterial and antifungal effects.
Fabrication of bimetallic (Mg-V)-doped TiO2 nanostructure via microwave-assisted sol-gel approach for enhanced electrocatalytic detection and photocatalytic degradation of hazardous tetracycline Samuel Chufamo Jikamo, P. Shyamala, Singupilla Sai Supriya, Sandhya Rani Nayak, NageswaraRao Kadiyala, Winnie Teja Dokka Next Materials, 2026 Nanocatalysts have emerged as powerful materials for environmental remediation, enabling both detection and removal of pollutants. In this study, magnesium and vanadium co-doped TiO 2 nanocatalysts were synthesized using a microwave assisted sol gel method and for the first time, employed as dual functional material for both the electrocatalytic detection and photocatalytic degradation of tetracycline in aqueous solution. The fabricated Mg-V co-doped TiO2 nanoparticles were characterized using various analytical techniques. The effects of dopant weight concentration and microwave irradiation power were systematically optimized, with the catalyst prepared at 0.25 wt% Mg and 1 wt% V under 540 W irradiation (MVT5M3) showed a lower band gap, smaller particle size, suppressed electron hole recombination than others. For electrochemical sensing, MVT5M3 was coated onto glassy carbon electrode (GCE) and characterized by cyclic voltammetry (CV) techniques. The catalyst enhanced sensitivity to 4.31 µA μM⁻¹ cm⁻² and a lower limit of detection (LOD) 0.3 µM for tetracycline detection using linear sweep voltammetry (LSV) techniques. In addition, the MVT5M3 catalyst achieved 93.53% photocatalytic degradation efficiency of tetracycline within 60 min under visible light irradiation. These results underline the potential of the Mg-V co-doped TiO 2 nanocatalyst as a highly efficient and bifunctional material for pharmaceutical pollutant monitoring and removal.

Forced Degradation Studies of Dobutamine and Isolation, Identification, and Characterization of Potential Degradation Impurity by Preparative HPLC, LC–MS/MS. Development and Validation of Stability Indicative UPLC Method Babu Reddy Goguladinne, Pulipaka Shyamala, K. M. V. Narayana Rao Biomedical Chromatography, 2025 Dobutamine (DB) is a β1‐adrenergic agonist with weak β2 activity and α1 selective activity, which is used clinically in cases of cardiogenic shock. Most of the available literature methods for DB that are LCMS incompatible have limitations for the identification of impurities. The current study describes forced degradation studies of DB and the identification of impurities by mass analysis, assigning structure to the major degradant, and the development and validation of an LCMS‐compatible UPLC method. Impurity A (dopamine) highly polar impurity listed in the dopamine US Pharmacopeial Monograph, was retained in the developed method without an ion pair reagent. The method has been developed using Waters Acquity UPLC system equipped with Acquity UPLC HSS PFP column (2.1 mm × 100 mm, 1.8 μ), 0.05% TFA in water:ACN (95:5 %v/v) as Mobile Phase A, and 0.05% TFA in water:ACN (5:95 %v/v) as Mobile Phase B using a gradient program. DB degradation behavior was studied by subjecting it to acidic, basic, neutral, and oxidative conditions as per ICH guideline Q1A (R2). One of the major degradants was isolated by preparative HPLC and identified characterization by LCMS/MS and HRMS. The newly developed UPLC method was found to be specific with respect to degradation products, and the method has been validated according to the ICH guidelines.
Structural Modification and Performance Enhancement of Zirconium Oxide with Copper Doping for Photo Catalytic and Antibacterial Applications J. Suresh Kumar, P. Shyamala, S. Gouthamsri, M. Ramanaiah Research Journal of Chemistry and Environment, 2025 Zirconium oxide (ZrO2) and copper-doped zirconium oxide (Cu-ZrO2) nanoparticles (NPs) were synthesized via a sol-gel combustion method to study the influence of doping on their photocatalytic and antimicrobial properties. X-ray diffraction (XRD) analysis confirmed a mixed tetragonal and monoclinic phase structure with decreasing crystallite size at higher dopant concentrations. The NPs were characterized using FTIR, XRD, SEM-EDX, XPS and UV-Vis spectroscopy. SEM revealed spherical morphology with minimal agglomeration. Under visible light, Cu-ZrO2 NPs exhibited enhanced photo- catalytic activity, degrading 96% of malachite green (MG) dye within 120 min. Antimicrobial assays demonstrated superior activity of Cu-ZrO2 against S.aureus, with an inhibition zone of 8 mm compared to E.coli.
Efficient Removal of Methylene Blue and Inactivation of Pathogens by Cobalt Doped ZrO2 Nanoparticles J. Suresh Kumar, P. Shyamala, S. Gouthamsri, M. Ramanaiah Ecs Journal of Solid State Science and Technology, 2025 In this investigation, we synthesized a pure ZrO2 and Co-doped ZrO2 nanoparticles, through glycine assisted sol-gel method. The phase purity of prepared nanoparticles was conformed through X-ray diffraction (XRD) and Fourier Transform Infrared (FTIR) techniques. The morphology and average crystallite size of pure and Co-doped ZrO2 nanoparticles was investigated using scanning electron microscopy (SEM), and observed average crystallite size were 24 (Co-doped ZrO2) and 20 nm (pure ZrO2). The Brunauer–Emmett–Teller (BET) technique was employed to study the surface area of the prepared nanoparticles, and observed surface area were 103 m2 g−1 (pure ZrO2) and 140 m2 g−1 (Co-doped ZrO2). High photocatalytic efficiency was observed in Co-doped ZrO2 nanoparticle due to improved surface area. The degradation of MB dye under visible light irradiation at various pH levels was also tested using the synthesised nanoparticles’ photo-catalytic capability. The prepared catalyst was the most effective for inactivation of S. aureus and may be utilized at large scale in treat wastewater effectively. It is noteworthy that the photocatalyst exhibits stability and reusability across several cycles. The work offers important new information about Co-doped ZrO2 nanoparticle potential as an effective antimicrobial agents and an effective photocatalyst for environmental clean-up.
Stereoselective reduction of enantiopure α-l-amino-β-keto esters using oxazaborolidine catalysts for the synthesis of statine analogues Malapati Venkateswara Reddy, Shyamala Pulipaka, Arvind Kumar Sharma Organic Communications, 2025 An efficient diastereoselective synthesis of Boc-protected 4-amino 3-hydroxy esters starting from natural amino acids is described.The key synthetic strategy involves diastereoselective reduction of -L-amino--keto esters in the presence of enantiopure R and S-2-Methyl-CBS-oxazaborolidine catalysts using borane as hydride source.The product diastereoselectivity depends upon the use of (R) or (S) enantiomer of 2-Methyl-CBS-oxazaborolidine.A reasonable mechanism is included which explains the diastereoselectivity of the reactions with the use of different enantiomers of 2-Methyl-CBS-oxazaborolidine.Furthermore, the resulting diastereomeric mixture of the reduced products can be separated by column chromatography to gain access to the single pure diastereomers.
