  • Subchronic Oral Exposure to Chlorpyrifos Decreases Butyrylcholinesterase in Wistar Rat

    Background: Indonesia as an agricultural country has more than 33 million workers in the agriculture. Pesticides have an important role in agriculture because they function as powerful plant pest exterminators. However, its use is considered to cause various toxic effects, one of which is on the nervous system. This study aimed to evaluate the impact of pesticide exposure on cognitive impairment by assessing the butyrylcholinesterase biomarker in the blood of rats as experimental animals.Methods: This was a true experimental study conducted during September–November 2022 at the Pharmacology and Biochemistry Laboratory, Faculty of Medicine, University of Jember with a post-test only approach using Wistar rats which were divided into 1 control group and 4 treatment groups treated with chlorpyrifos 5 mg/kg orally. Serum butyrylcholinesterase enzyme levels were measured using the kinetic photometric method and then analyzed with a regression test in IBM SPSS® Statistics software. Results: The average value of butyrylcholinesterase was 920.23±145.27 (Control group); 904.48±91.6 (7 days after chlorpyrifos administration); 889.81±95.2 (14 days after chlorpyrifos administration); 457.84±69.5 (28 days after chlorpyrifos administration); 575.82±194.25 (56 days after chlorpyrifos administration). The results of the One-Way ANOVA test showed significant differences between groups as indicated by a significance test of 0.024 (p <0.05).Conclusions: There is a relationship between the duration of exposure to chlorpyrifos and butyrylcholinesterase levels in Wistar rats. This fact shows that the longer an organism is exposed to low doses of chlorpyrifos pesticides, the higher the level of neurological system toxicity that occurs in that organism.
  • Gel of Monascus purpureus JmbA rice accelerate wound healing in diabetic rats

    The most frequent complication of diabetes mellitus (DM) is diabetic ulcer, with a prevalence of 15-20% and an amputation rate of 30%. There is no effective and easy-to-apply treatment for acute-phase open wounds of patients with DM to prevent their development into ulcers. Monascus purpureus jmbA Rice (MJR) from Jember is superior to similar products from other regions in Indonesia because it has the highest monacolin content with an antioxidant effect. The study aimed to prove the effectiveness of MJR gel in accelerating the healing of acute wounds of diabetic rats. This true experimental laboratory research was a posttest-only control group design. We used Rattus norvegicus Wistar rats, 3-4 months old, weighing 200-300 grams. The 36 rats were divided into four treatment groups. The control group was given a gel of MJR 0%. The positive control group was given the antibiotic and placenta extract gel combination, and treatment groups T1 and T2 were given gel MJR40% and MJR80%. Excision wounds were made as deep as the subcutaneous layer on the back of the hyperglycemic rat. Four rats were terminated from each group on days 4, 11, and 18, and skin tissue was taken to observe the percentage of wound area shrinkage, epithelial thickness, and fibroblast count and analyze the levels of hydroxyproline and MDA. The MJR80% group had the highest percentage of wound area shrinkage, epithelial thickness, and fibroblast count compared to other groups. MDA levels decreased with increasing days in all treatment groups. Hydroxyproline levels increased on days 4 and 11, then decreased on day 18 in all treatment groups. We conclude that the 80% MJR group revealed the best acute diabetic wound healing. It was not statistically different from the combination of antibiotic and placenta extract gel treatment group.
  • The effect of chlorpyrifos oral exposure on the histomorphometric and kidney function in Wistar rat

    BACKGROUND: Chlorpyrifos belongs to a broad-spectrum organophosphate insecticide that has high toxicity, is metabolized in the liver by the oxidation reaction, and can inhibit acetylcholinesterase activity. Acetylcholinesterase inhibition generates the reactive oxygen species and induces oxidative stress, which ultimately results in cellular damage like in the kidney. Examining blood urea nitrogen (BUN) levels, creatinine, and kidney histopathology is an appropriate indicator to assess the toxicity of chlorpyrifos to the degree of damage to cells and kidney tissue. MATERIALS AND METHODS: This research used to determine the effect of duration of exposure to chlorpyrifos and dose–response relationships is important for early detection of the effects of chlorpyrifos toxicity on health. The research study was a true experimental (completely randomized design) consisting of 30 subjects divided into 5 groups. Controlled Group (K1) given 1 mg/kg BW Tween 20 and NaCl 0, 9% until the 56th day. The chlorpyrifos exposed group (P1, P2, P3, and P4) was given chlorpyrifos 5 mg/kg BW for 7, 14, 28, and 56 days. After the treatment, BUN and creatinine levels were measured, and microscopic changes in the kidney were analyzed. The results of BUN, creatinine, and kidney histopathologic were analyzed using the analysis of variance statistical test. RESULTS: The data result showed that compared to the control group, there were significant increases of BUN and creatinine (P = 0.013 and P = 0.003). Histopathological examinations of kidney glomerulus diameter were also smaller compared to the control group (P = 0.00). All the data measurement indicates significant differences compared to the control group. CONCLUSIONS: We concluded that sub-chronic oral exposure to chlorpyrifos at low doses can damage the kidneys and cause kidney failure.
  • Effectiveness of Edamame (Glycine max L. Merril) Membrane in Accelerating The Wound Healing Process of Deep-Partial Thickness Burn

    The gold standard for deep-partial thickness burns is early excision and skin graft; however, many hospitals in Indonesia still use conventional treatment due to the high cost of surgery and the requirement of qualified medical professionals. This research aimed to study the effectiveness of edamame (Glycine max. L Merill) membrane as therapeutic innovation in deep-partial thickness burns. Forty-eight male Wistar rats with deep-partial thickness burns were assigned randomly to four groups, including control and treatment (silver sulfadiazine, the membrane with 40% and 60% edamame extract). Measuring wound healing parameters such as macroscopic evaluation, histopathologic, and hydroxyproline was examined on days 4, 10, and 16. Treatment groups of membrane edamame significantly improved wound healing than the control group. Macroscopically, histopathological findings and hydroxyproline assay confirmed the efficacy of the edamame membrane at 60%, which provided the best healing results. This study showed that edamame membrane is effective as deep-partial thickness burns wound dressing.

  • Kefir stimulates anti-inflammatory response in TB-AFB (+) patients

    Regular consumption of kefir promotes beneficial physiological and therapeutic effects including stimulation of immune response. In this study, we investigated the immunomodulatory effects of kefir on peripheral mononuclear cells (PBMCs) derived from patients diagnosed with pulmonary tuberculosis (TB). PBMCs were isolated, treated with different concentration of kefir (1/20, 1/50, 1/100 and 1/200) and cultured for 4 days. The proportion of CD4+ and CD8+ cells was analyzed by flow cytometry and the production of Th1 cytokines IL-2 and IFN-γ and Th2 cytokines IL-4 and IL-10 was determined by ELISA. The results indicate that kefir slightly stimulated CD4+ as well as CD8+, but the effect was insignificant. Low doses of kefir significantly induced the secretion of IL-10 but not IL-2, IL-4, or IFN-γ. We conclude that kefir in a low concentration stimulates Th2 rather than Th1-type immune response and therefore works as an immunosuppressant for early diagnosed TB patients.
