  • Comparative Enzymatic and Gene Expression Responses in Wheat to DON- and NIV-Producing Fusarium Species

    Fusarium head blight (FHB) is a major threat to wheat production that is caused by toxigenic species of the Fusarium graminearum complex. This study aimed to investigate the biochemical and molecular defense responses of Brazilian wheat genotypes (BRS 194, BRS Parrudo, and Frontana) with contrasting FHB susceptibilities following inoculation with F. graminearum (deoxynivalenol producer) and F. meridionale (nivalenol producer). Temporal patterns of antioxidant enzymes, defense-related enzymes, and gene expression (ABC-Transporter and Ca2+-ATPase) were analyzed from 12 to 96 h after inoculation. The ANOVA results revealed significant effects of genotypes, inoculation, and time after inoculation on most of the evaluated enzymatic activities. Frontana exhibited high basal activity for most enzymes, and after inoculation, the enzyme activity was higher than in other genotypes. BRS 194 presented delayed and fragmented activation patterns, particularly under DON-producing pathogen infection. According to the transcriptome results, inoculation with the NIV-producing pathogen upregulated both genes, reaching up to an 18-fold increase. BRS 194 showed an upregulated transcript pattern from the early hours after inoculation. Frontana showed increased transcript levels, reaching 12-fold, under DON-producing pathogen infection. These findings show that biochemical and molecular responses varied depending on genotype and the chemotype of the Fusarium isolate, highlighting the importance of early, coordinated defense activation in FHB resistance.
  • Comparative analysis of selected nutrients in Pereskia species

    This investigation undertakes a comparative quantitative assessment of the macronutrient and micronutrient profiles of Pereskia grandifolia (Haworth) and Pereskia aculeata (Miller), commonly known as Ora-pro-nóbis, categorized as Non-Conventional Food Plants (NCFPs), juxtaposing these profiles against established food crops. Quantitative analysis revealed that both Pereskia species exhibit elevated concentrations of micronutrients such as iron and zinc, surpassing those of Phaseolus vulgaris (Linnaeus) and Lens culinaris (Linnaeus). Specifically, solubilized zinc levels in P. aculeata (13.6 mg/100g) and P. grandifolia (13.9 mg/100g) exceed those of lentils (7.5 mg/100g) and beets (3.7 mg/100g). Similarly, solubilized iron levels in P. grandifolia (12.4 mg/100g) and P. aculeata (10.5 mg/100g) surpass those of lentils (3.0 mg/100g) and beets (3.8 mg/100g). However, phytate levels are comparatively lower in the Pereskia species, with P. grandifolia demonstrating reduced concentrations at 1500 mg/kg, alongside significant preponderance in protein (20.8 g/100g) and nitrogen (14.6 g/100g). Principal Component Analysis (PCA) was applied, demonstrating a strong associative propensity of iron, zinc, and protein with the Pereskia genus. In contrast, Pearson's correlation coefficients elucidated a complex interdependency among the nutrient variables, except for an inverse correlation between phytate concentration and both iron and protein levels. In conclusion, this research sheds light on the enriched nutritional profiles of P. grandifolia and P. aculeata, emphasizing their potential as valuable dietary resources.
  • Biological dynamics of no-tillage soils in the western region of Paraná

    This study investigates soil dynamics on farms in the western region of Paraná, Brazil, highlighting the importance of biological parameters in agriculture. In particular, focusing on the interaction of management practices with soil biodiversity and biological functions, the aim is to understand and promote sustainable and efficient agricultural practices. To do this, we collected soil samples from 15 farms close to Toledo, Paraná, Brazil. These samples were then analyzed to determine biological and physicochemical parameters using techniques such as carbon and nitrogen microbial biomass, metabolic coefficient, basal respiration, bacterial and fungal biomass, and length of the hyphae. The most contrasting soils were evaluated for physicochemical composition and metagenomic analyses. The results showed significant differences in biological parameters between 2020 and 2021, including fungal biomass, hyphae length, and soil basal respiration. Statistical analyzes revealed strong relationships between biological variables, notably the correlation between fungal hyphae and total nitrogen. Climate changes and management practices appear to influence the microbial composition and biological functions of the soil over the years. Soil P9 stood out with superior biological activity and richer microbial diversity, contrasting with soil P13. These differences reflect the influence of management and climatic conditions on soil composition and biological functions. The microbial comparison of the soils emphasized the need for continuous and careful agricultural management, highlighting the importance of biodiversity and ecological functionality of the soil for agricultural sustainability. So, the study underscores the relevance of considering soil biological parameters, in addition to physicochemical aspects, to optimize soil health and productivity.

    Pseudomonas fluorescens, a Gram-negative bacterium abundant in soil, plays a critical role in promoting plant growth and controlling pathogens, demonstrating remarkable biocontrol capabilities. This review explores the utility of P. fluorescens and its ability to produce secondary metabolites, IAA (Indole-3-acetic acid), and siderophores, addressing agricultural challenges under the strains of climate change. It emphasizes its role as a plant growth-promoting bacterium (PGPB), synthesizing recent findings on its contributions to enhancing plant resilience, pathogen resistance, and sustainable agricultural practices. The production of secondary metabolites, IAA, and siderophores by P. fluorescens is examined for its effectiveness in biocontrol, nutrient mobilization, and hormonal regulation. These functions are critically analyzed through diverse research methodologies, including laboratory and field trials, underscoring the bacterium’s pivotal role in advancing agricultural sustainability and productivity. As the agricultural sector increasingly focuses on bio-products and the exploration of soil microorganisms, P. fluorescens emerges as a promising solution to enhance farming resilience in the face of climatic adversities.
  • Hydroponic effluent as an alternative fertilizer in bell pepper production

    As hydroponic systems expand and generate effluents, research has increasingly focused on their management due to potential environmental impacts. This study examined the effectiveness of using hydroponic effluents as fertilizer in bell pepper cultivation. The chemical composition of the effluents was analyzed and compared against water quality standards set by Resolution No. 222/02 of SEAM. The study employed two application methods (foliar and root) and three concentrations (0%, 50%, 100%) of effluents. A randomized complete block design was used to evaluate plant height, fruit number, and yield. Results showed that a 50% foliar application significantly increased plant height compared to the 100% concentration, with no significant differences in fruit number and yield. For root application, no significant differences were observed across the variables at different concentrations, although the best performance was noted at 50%. Comparative analysis between application methods revealed that root application resulted in taller plants but lower productivity. The study concludes that using hydroponic effluents as fertilizers affects bell pepper productivity, with a 50% concentration being most effective regardless of the application method.

    Biological nitrogen fixation (BNF) by diazotrophic bacteria is one of the oldest and most crucial processes in nature. In this process, bacteria form symbiotic associations with plants, capturing atmospheric nitrogen and making it readily available to them. The diversity of nitrogen-fixing bacteria is vast. Recent advancements in molecular biology techniques have enabled the identification of new genera and species capable of fixing nitrogen and providing other types of nutrients for plants. From an agronomic perspective, this process is fundamental in increasing crop productivity sustainably and -cost-effectively. This review aims to categorize the most recent updates on the diversity of nitrogen-fixing bacteria and showcase the main advances in the genetic improvement of legumes for this characteristic. Recent research has revealed a wide diversity of species applicable to various crops of agronomic interest, and many of these bacteria have been used either alone or in consortium with other microorganisms. This study demonstrates the agricultural potential of these new discoveries and the vast possibilities for expanding research into the diversity of microorganisms responsible for BNF in agriculture.

    The soil is a dynamic environment, influenced by abiotic and biotic factors, which can result in changes in plant development. This study aimed to assess the impact on vegetative growth of chia (Salvia hispanica L) inoculated with Trichoderma harzianum and on the rhizosphere microbiome. The experimentation was conducted in a greenhouse under controlled conditions growing chia plants in pots containing soil with a clayey texture. Different concentrations of T. harzianum (0; 2.5; 5.0; 10.0; 20.0 µL. g-1 of seed) were applied to the chia seeds before planting. Morphological parameters, including plant height (cm), number of branches, stem diameter (mm), number of days to flowering and shoot and root dry masses (g) were quantitatively assessed. After the cultivation period, soil samples from the rhizosphere region were collected for subsequent chemical and metagenomic analyses. These samples were also compared with the control soil, collected before installing the experiment. The results showed that increasing doses of T. harzianum promoted a significant increase in the diameter of the stem, number of branches, dry biomass of the root system and the number of days to flowering, without modifying the overall height of the plants. Soil metagenomics indicated that T. harzianum inoculation modified the microbial diversity of the rhizosphere environment, with more pronounced effects observed in samples treated with higher concentrations of the inoculant. Furthermore, there were changes in the chemical composition and enzymes related to soil quality in correlation with the concentrations of the applied inoculant. This study demonstrated that inoculating chia seeds with T. harzianum not only promotes specific morphogenetic characteristics of the plant, but it also has a significant impact on the microbial diversity and biochemical functionality of the soil, including an observed increase in the populations of T. harzianum and T. asperellum.
