Badino International Journal of Chemical Reactor Engineering, 2023 The hydrodynamics of airlift bioreactors, which offer an interesting alternative to conventional stirred-tank bioreactors, has generally been evaluated using experimental approaches, requiring time, energy, and reagents. However, computational fluid dynamics (CFD) has emerged as an important and valuable tool for the analysis and design of these devices, saving time and experimental effort, while providing a large amount of information. In this study, four geometries of a square cross-section 10-L split airlift bioreactor operating with distilled water were simulated using CFD, and the hydrodynamics variables gas hold-up and liquid velocity were evaluated. CFD satisfactorily predicted the hydrodynamic parameters, when compared to experimental data, allowing adequate prediction of the shear rate distribution in airlift bioreactors. The results indicated that different shear rate distributions were obtained by geometric modifications in the bioreactor, showing that its design should be considered to satisfy different specific bioprocess requirements.
Synergistic Effects of Stirring and Aeration Rate on Carotenoid Production in Yeast Rhodotorula toruloides CCT 7815 Envisioning Their Application as Soap Additives Renata M. M. G. P. Fermentation, 2023 The production of carotenoids by microbial organisms has gained significant interest due to the growing demand for natural products. Among the non-model oleaginous red yeasts, Rhodotorula toruloides stands out as an appealing host for natural carotenoid production. R. toruloides possesses the natural ability to metabolize a wide range of substrates, including lignocellulosic hydrolysates, and convert them into lipids and carotenoids. In this study, we focused on utilizing xylose, the main component of hemicellulose, as the major substrate for R. toruloides. We conducted a comprehensive kinetic evaluation to examine the impact of aeration and agitation on carotenoid production. Results in stirred-tank reactor demonstrated that under milder conditions (300 rpm and 0.5 vvm), R. toruloides accumulated over 70% of its cell mass as lipids. Furthermore, the highest carotenoid yields were achieved at high agitation rates (700 rpm), with carotenoid levels reaching nearly 120 µg/mL. Several carotenoids were identified, including β-carotene, γ-carotene, torularhodin, and torulene, with β-carotene being the major carotenoid, accounting for up to 70% of the total carotenoid content. The carotenoid-rich extract produced by R. toruloides under evaluated conditions was successfully incorporated into soap formulations, demonstrating the addition of antioxidant properties. This work provides a comprehensive understanding of xylose conversion into natural carotenoids by R. toruloides, presenting a promising avenue for their application in cosmetics. Furthermore, this study highlights the potential of a renewable and cost-effective approach for carotenoid production in the soap industry.
Solvent-Free Enzymatic Synthesis of Dietary Triacylglycerols from Cottonseed Oil in a Fluidized Bed Reactor Daniela Remonatto, Núbia Santaella, Lindomar Alberto Lerin, Juliana Cristina Bassan, Marcel Otávio Cerri, et al. Molecules, 2023 The synthesis of structured lipids with nutraceutical applications, such as medium-long-medium (MLM) triacylglycerols, via modification of oils and fats represents a challenge for the food industry. This study aimed to synthesize MLM-type dietary triacylglycerols by enzymatic acidolysis of cottonseed oil and capric acid (C10) catalyzed by Lipozyme RM IM (lipase from Rhizomucor miehei) in a fluidized bed reactor (FBR). After chemical characterization of the feedstock and hydrodynamic characterization of the reactor, a 22 central composite rotatable design was used to optimize capric acid incorporation. The independent variables were cycle number (20–70) and cottonseed oil/capric acid molar ratio (1:2–1:4). The temperature was set at 45 °C. The best conditions, namely a 1:4 oil/acid molar ratio and 80 cycles (17.34 h), provided a degree of incorporation of about 40 mol%, as shown by compositional analysis of the modified oil. Lipozyme RM IM showed good operational stability (kd = 2.72 × 10−4 h−1, t1/2 = 2545.78 h), confirming the good reuse capacity of the enzyme in the acidolysis of cottonseed oil with capric acid. It is concluded that an FBR configuration is a promising alternative for the enzymatic synthesis of MLM triacylglycerols.
