Cactus Cladodes and Sugarcane Bagasse Can Partially Replace Earless Corn Silage in Diets of Lactating Dairy Cows Izaac P. S. Medeiros, Sebastião I. Guido, Marco A. S. Gama, Carlos H. M. Dairy, 2024 This study aimed to evaluate the effects of replacing earless corn silage (ECS) with cactus cladodes (CC; Opuntia spp.) and sugarcane bagasse (SB) on nutrient intake, digestibility, feeding behavior, milk yield (MY), and composition of lactating dairy cows. Ten Holstein cows, weighing 571 ± 97.0 kg and producing 23.0 ± 4.4 kg of milk per day, were assigned to two contemporaneous 5 × 5 Latin squares. Treatments consisted of five levels of ECS replacement with CC plus SB (0, 25, 50, 75, and 100%). The results showed a linear increase in dry matter (DM) intake (p < 0.05) (15.98 and 18.73 kg/day) and a quadratic increase (p < 0.05) in crude protein and energy intake (2.97 kg/day and 27.52 Mcal/day at 95.4 and 88.6% substitution, respectively). Apparent DM digestibility increased (p < 0.05), but fiber digestibility decreased linearly (p < 0.05). Treatments had a quadratic effect (p < 0.05) on MY and fat-corrected MY (24.17 kg/day and 21.9 kg/day at 63.9% and 38.6% CC plus SB, respectively). Milk fat (3.26 and 2.35%) and total solids content decreased linearly (p < 0.05), whereas the percentages of protein, lactose, and nonfat solids increased (p < 0.05). Additionally, the CC–SB diets linearly reduced the time spent on feeding and rumination and total chewing time. For Holstein cows fed common semiarid diets, milk production can be maximized by replacing 38.6% of ECS with CC plus SB.
Phenotypic variation in milk fatty acid composition and its association with stearoyl-CoA desaturase 1 (SCD1) gene polymorphisms in Gir cows Carolina Guimarães Ramos Matosinho, Pablo Augusto de Souza Fonseca, Maria Gabriela Campolina Diniz Peixoto, Izinara Cruz Rosse, Fernando César Ferraz Lopes, et al. Journal of Animal Breeding and Genetics, 2023 Individual variation in milk fatty acid (FA) composition has been partially attributed to stearoyl-CoA desaturase 1 (SCD1) gene polymorphisms in taurine breeds, but much less is known for Zebu breeds. This study investigated the phenotypic variation in milk FA composition, and the influence of SCD1 variants on this trait and on milk fat desaturase indices (DI) in Gir cows. The functional impact of SCD1 variants was predicted using bioinformatics tools. Milk and blood samples were collected from 312 cows distributed in 10 herds from five states of Brazil. SCD1 variants were identified through target sequencing, and milk FA composition was determined by gas chromatography. Phenotypic variation in milk FA composition fell within the range reported for taurine breeds, with SCD18 index showing the lowest variation among the DI. Fourteen SCD1 variants were identified, six of which not previously described. Regarding the A293V polymorphism, all cows were homozygous for the C allele (coding for alanine), whereas all genotypes were detected for the second SNP affecting the 293 codon (G > A), with compelling evidence for functional effects. Significant associations (based on raw p-values) were found between this SNP and C12:0, cis-9, trans-11 CLA and short-chain FA, and between another SNP (rs523411937) and C15:0 and odd-chain linear FA. A new SNP on Chr26:21277069 was associated with trans-11 C18:1, cis-9, trans-11 CLA, C18:3 n-3 and n-3 FA. These findings indicate that SCD1 polymorphisms also contributes to the phenotypic variation in milk FA composition of Gir cows, with potential use in their breeding programmes.
Nutritional Quality of Milk Fat from Cows Fed Full-Fat Corn Germ in Diets Containing Cactus Opuntia and Sugarcane Bagasse as Forage Sources Camila S. da Silva, Marco A. S. Gama, Erick A. M. Animals, 2023 We evaluated the performance, milk composition, and milk fatty acid profile of cows fed diets composed of cactus cladodes (Opuntia stricta [Haw.] Haw), sugarcane bagasse and increasing levels of full-fat corn germ (FFCG). We hypothesized that ground corn can be effectively replaced by FFCG when cactus cladodes and sugarcane bagasse are used as forage sources. The cows were randomly distributed into two 5 × 5 Latin Squares and fed five diets in which ground corn was progressively replaced with full-fat corn germ (FFCG; 0%, 25%, 50%, 75%, or 100% of substitution). Adding FFCG to the diet increased milk production and milk fat content and reduced milk protein content. Overall, FFCG reduced the proportion of saturated FAs and increased mono- and polyunsaturated FAs in milk, including CLA isomers. In addition, activity indices of stearoyl-CoA desaturase were reduced by increasing levels of FFCG. We conclude that the substitution of corn for FFCG in diets based on cactus cladodes and sugarcane bagasse positively modifies the FA profile of milk and could add commercial value to milk products (e.g., CLA-enriched milk). In addition, the milk fat response indicates that the basal diet was favorable to the rumen environment, preventing the trans-10 shift commonly associated with milk fat depression.

RAPID METHOD FOR SIMULTANEOUS DETERMINATION OF CIS-9, TRANS-11 AND TRANS-10, CIS-12 CONJUGATED LINOLEIC ACID ISOMERS IN MILK BY GC-FID Brenda Porto, Isaura Faria, Jéssica de Souza, Olívia Moreira, Marco da Gama, et al. Quimica Nova, 2023 Ruminant milk is the lead source of conjugated linoleic acid (CLA) in the human diet, with cis-9, trans-11 CLA being the major among all. Small amounts of trans-10, cis-12 CLA are also found in synthetic supplements. Since both isomers are biologically active with potential health benefits, there is great interest in quantifying them for quality control routines. An alternative method for the analysis of the aforementioned CLAs by fast gas chromatography (GC) is discussed in the present study. The fatty acid methyl ester mixture obtained by alkaline catalysis was injected into a GC equipped with a flame ionization detector (FID) and fitted with an ionic liquid SLB-IL111 chromatographic column (15 m × 0.10 mm × 0.08 µm). Separation was achieved in less than 5 min using a 168 °C isotherm run. Both CLA isomers were quantified by using of single point standard addition statistical approach. Results were contested to those obtained using the recommended 100 m long CP-Sil88 capillary column and none evidence of significant differences was found within 95% confidence interval. Therefore, the proposed method could be valuable to focused regulatory routines of large numbers of samples with greater analytical frequency.
