Background: Current pharmacological treatments for acute orofacial pain present limitations, risks, and side effects. Thus, the search for safer alternatives is justified. The essential oil of Alpinia zerumbet (EOAz) has been used in the treatment of several medical conditions, including pain and inflammation, but scientific validation is scarce. The aim of this study was to investigate the antinociceptive and anti-inflammatory effects of EOAz in models of acute orofacial pain in rats and in inflammatory parameters in vitro. Methods: Adult male Wistar rats were subjected to intraoral incision surgery, a model of postoperative pain. The effect of the facial topical application (1 to 3 times a day for 3 days) of EOAz or a formulation based on EOAz (Fb-EOAz) was assessed on heat and mechanical hyperalgesia. The same rats were tested in the open field test (OFT) to assess the influence of the treatments on rats’ locomotion. Moreover, the effects of the treatments were evaluated on facial heat hyperalgesia induced by lipopolysaccharide (LPS), a model of myofascial pain, and in vitro, in the release of nitric oxide (NO) and interleukin-6 in macrophages stimulated by LPS. Results: EOAz and Fb-EOAz reduced postoperative heat hyperalgesia, but only Fb-EOAz reduced postoperative mechanical hyperalgesia and heat hyperalgesia induced by LPS. Fb-EOAz reduced NO and interleukin-6 release by macrophages stimulated by LPS. None of the treatments affected the rat’s locomotion. Conclusions: These data provide preclinical evidence of antinociceptive and anti-inflammatory effects of Fb-EOAz. This approach may represent an alternative or adjuvant therapy in the control of inflammatory and myofascial orofacial pain. - In Vitro Assessment of the Effects of Endodontic Sealers on Cell Viability, Adhesion, and Migration

This study aimed to evaluate the in vitro cytocompatibility and bioactivity of the newly launched calcium silicate‐based sealer BioRoot Flow, compared with other calcium silicate‐based sealers (AH Plus Bioceramic Sealer and BioRoot RCS) and the epoxy resin‐based AH Plus Jet. NIH/3 T3 fibroblasts were used to assess cell viability, adhesion, and migration. Material characterisation was performed using SEM, EDS, and XRD. BioRoot RCS exhibited the highest cell viability, while BioRoot Flow showed fewer adherent cells and reduced migration at 72 h. EDS revealed zirconium content in BioRoot Flow, and XRD confirmed calcite presence in all calcium silicate‐based sealers. Variations in cell behaviour may be related to differences in chemical composition and surface properties, highlighting the need for further investigation. - Effect of parecoxib and dexamethasone on the temporomandibular joint of orchiectomized rats: Morphological and immunological analysis

Background/Objectives: Regenerating critical-sized bone defects is a significant clinical challenge. Autogenous bone grafts are the gold standard but have limitations, including donor site morbidity. As an alternative, this study introduces a novel biocomposite combining an ethyl cyanoacrylate (ECA) polymer with Hancornia speciosa (Hs) latex. The ECA acts as a scaffold and delivery vehicle for the latex, which contains phytochemicals with known angiogenic properties. Methods: We created 5 mm critical-sized calvarial defects in 36 Wistar rats, which were divided into four experimental groups. Bone regeneration was evaluated at 30, 60, and 90 days using micro-computed tomography (micro-CT) for morphometric analysis and hematoxylin and eosin staining for histology. Results: The composite-treated group (Hs+ECA) showed significantly higher bone volume (57.2; IQR: 56.7–61.2) than the control (53.9; IQR: 49.4–56.4) and ECA-only (48.4; IQR: 47.2–59.9) groups at 90 days (p < 0.05). By day 60, the bone volume in the Hs+ECA group was statistically similar (p > 0.05) to that of the autogenous bone group. Histological analysis revealed an organized repair process with neoangiogenesis observed only in the Hs+ECA group, confirming the material’s strong bioactivity. Conclusions: The Hs+ECA composite is a promising biomaterial that acts as an effective delivery system for the bioactive components of the latex. The induced angiogenesis was critical to its regenerative success. Background: Acupuncture is a traditional Chinese medicine technique that involves the insertion of filiform needles into the body. Currently, various brands of acupuncture needles are available for purchase and product quality control is intended to ensure the safety of both the patient and the acupuncturist. Objective: The purpose of this study was to assess the quality of acupuncture needles using physical–chemical and mechanical tests according to International Organization for Standardization (ISO) standard 17218:2014. Methods: We evaluated nine different brands of 0.25 mm × 30 mm acupuncture needles. The needles were characterized using scanning electron microscopy—energy dispersive X-ray spectroscopy (SEM-EDS) techniques for morphological and compositional evaluation at 300× magnification. Mechanical tests were used to determine puncture performance, intensity and needle tip sharpness according to ISO 17218:2014 using an Instron 4411 with controlled needle displacement. Puncture force was measured in Newtons, and a stereomicroscope was used to evaluate structural changes after the intensity test. Results: SEM-EDS analyses indicated deformations, scratches and imperfections in various brands of needle, particularly Açomed and Complementar. Following the intensity test, all brands exhibited deformations at the needle tips. The Uniqmed brand had the lowest puncture performance test force (median = 0.033 N, 95% confidence interval (CI) = 0.030–0.035 N), while Açomed had the highest (0.117 N, 95% CI = 0.109–0.132 N). The Dragon brand had the lowest sharpness test force (0.026 N, 95% CI = 0.025–0.029 N), while Complementar had the highest (0.060 N, 95% CI = 0.056–0.064 N). Conclusion: Despite statistically significant disparities, no group exceeded the limits set by ISO 17218:2014. Although there are a few research studies on this topic, some authors have opted not to reveal brand names, which makes direct comparisons problematic. AimCalcium silicate‐based cements have been widely used in dentistry mainly due to their physicochemical and biological properties. Commercially available materials use radiopacifiers containing metals (bismuth, tantalum, tungsten and/or zirconium).To investigate volumetric changes, in vivo biocompatibility and systemic migration from eight commercially available materials, including powder/liquid and ‘ready‐to‐use’ presentations.MethodologyAfter characterization, tubes were implanted in healthy Wistar rats' alveolar bone and subcutaneous tissues. Micro‐CT was used to evaluate volumetric change before/after 30 days of implantation. Histological and immunohistochemistry analysis were used to evaluate materials' biocompatibility. After euthanasia, kidney samples were retrieved, acidic digested and evaluated by inductively coupled plasma mass spectrometry (ICP‐MS) for bismuth, tantalum, tungsten and zirconium mass fractions. Statistical analysis compared the results for normality and comparisons adopted a level of significance of 0.05.ResultsCharacterization photomicrographs and spectroscopy analysis revealed calcium, silicon and radiopacifiers for tested cements. Volumetric changes after implantation showed higher alteration in subcutaneous tissues than alveolar bone indicating that Biodentine, EndoSequence BC RRM Putty and ProRoot MTA were the most stable materials. Histological analysis found intense inflammation for NeoPUTTY and moderate for the other materials; osteocalcin and osteopontin were positively marked for all materials. Despite its volumetric stability, ProRoot MTA showed a 1000‐fold higher mass fraction of bismuth accumulation and MTA Repair HP a 37‐fold higher tungsten accumulation in kidney samples when compared with the nonexposed controls. All tantalum‐analysed samples indicated a similar mass fraction with the nonexposed controls. Biodentine exhibited a significant lower kidney mass fraction of zirconium accumulation when compared with this control.ConclusionsVolumetric analysis revealed that Bio‐C Repair, NeoPUTTY and MTA Repair HP presented greater volumetric loss when implanted in the subcutaneous tissue. NeoPUTTY presented more intense inflammatory infiltrate. Systemic migration analysis highlighted the predominance of bismuth in the ProRoot MTA group. These results suggest that endodontic repair cements are affected by their chemical composition, the type of implant tissue and different clinical settings. - Physicochemical properties and biological interaction of calcium silicate-based sealers - in vivo model
