Carbon Dot-Based Electrochemical and Optical Sensors for Pharmaceutical Analysis and Point-of-Care Diagnostics Ganesh Gollavelli, Chiranjib Patra, Chiranjeevi Korupalli, Manuri Brahmayya, Yong-Chen Ling Biosensors, 2026 Because of their special optical and electrochemical characteristics, superior biocompatibility, adjustable surface chemistry, and inexpensive, scalable synthesis, carbon dots (CDs), including carbon quantum dots and graphene quantum dots, have become powerful and adaptable nanomaterials for advanced pharmaceutical analysis and other toxicants. The sensitive and selective detection of active pharmaceutical substances, degradation products, contaminants, biomarkers, and therapeutic medication levels in complex matrices has shown great promise in recent years with CD-based nanobiosensors. The development of various sensing platforms, such as electrochemical, optical, and dual-mode biosensors, as well as integration into microfluidic, paper-based, and wearable point-of-care (POC) devices, is made possible by their intrinsic fluorescence, effective electron transfer capacity, and ease of functionalization. With an emphasis on sensing mechanisms, biorecognition techniques, and analytical performance, this study critically reviews current developments in CD-based nanobio/chemosensors for pharmaceutical analysis. It includes a thorough discussion of important applications in drug development, stability research, therapeutic drug monitoring, and drug quality control. Along with new developments like green synthesis, AI-assisted signal processing, and smart sensing platforms, current issues with reproducibility, standardization, biocompatibility, and regulatory validation are highlighted. Lastly, prospects for the industrial application and clinical translation of CD-based nanobiosensors are discussed.
Nano-sulfonated Graphene Oxide: A Metal-free Catalyst for the Synthesis of Benzimidazoles and Antimicrobial Activities V Murali Krishna Madasu, M Padma, Jyotsna Cherukuri, Kadali Yashoda, E Vijaya, CH Praveen, Manuri Brahmayya Journal of Environmental Nanotechnology, 2026 Nano sulfonated graphene oxide (SGO) was synthesized with slight modifications to previously reported methods. An efficient and ecofriendly protocol was developed for the synthesis of benzimidazole derivatives using o-phenylenediamine and D-glucose as substrates, employing a metal-free sulfonated graphene oxide carbocatalyst. A diverse range of benzimidazole derivatives was obtained in moderate to high yields (up to 94%) using a sustainable and readily available C1 source under sonication conditions. The reaction, carried out in ethanol with tertiary-butyl hydroperoxide, provides a general and novel approach for benzimidazole synthesis via a carbonylation pathway. The process is facilitated by a cost-effective and environmentally benign SGO nanocatalyst, enabling the formation of valuable products in excellent yields. The synthesized benzimidazole derivatives exhibited strong antibacterial activity, with inhibition rates of 99% to 100% against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Streptococcus pyogenes.
Sulfonated Graphene Oxide Catalyzed Transformation of Pyrazole-Oxazolidine Derivatives and their Antimicrobial Activities V. Murali Krishna Madasu, Manuri Brahmayya, S. Deepthi, A. Harinath, Lakshmi Rekha Buddiga, J. Chandra Sekhara Rao, M. Padma, Praveen Choppara, D. Samsonu, A. Venkateswara Rao Asian Journal of Chemistry, 2026 In this work, biologically active novel (E)-5-((1-phenyl-3-(4-R-phenyl)-1H-pyrazol-4-yl)methylene)-3-(4-chlorophenyl)oxazolidine-2,4-dione derivatives are synthesized and characterized. Initially, 5-azanyl(4-chlorophenyl)carbamate (3) was obtained from 4-chloroaniline (1) via carbonation with CO2 (2) in liquid ammonia, followed by O-carboxymethylation with sodium 2-chloroacetate to afford sodium 2-(((4-chlorophenyl)carbamoyl)oxy)acetate (4). Subsequently, compound 4 was cyclized to yield 3-(4-chlorophenyl)oxazolidine-2,4-dione (5) using sulfonated graphene oxide (SGO) as metal free nanocatalyst. Later, benzohydrazide (6) was condensed with various types of carboxylic acids (7) in presence of glacial acetic acid to obtain (E)-(4-R-phenyl)ethylidene hydrazone (8). It is further treated with POCl3 at 80 ºC for 4 h to obtain substituted pyrazole derivatives (9a-h). Finally, substituted pyrazole derivatives (9a-h) treated with sulfonated graphene oxide (SGO) catalyzed product 3-(4-chlorophenyl)oxazolidine-2,4-dione (5) to obtain targeted heterocyclic molecules 10a-h. This protocol demonstrates significant advantages in efficiency, ecological sustainability and catalyst reusability. Furthermore, the synthesized 1,3-oxazolidine-2,4-dione derivatives were evaluated for their antibacterial properties. The findings highlight the catalytic potential of SGO in the synthesis of 1,3-oxazolidine-2,4-dione derivatives as well as in broader applications in green organic transformations.
Synthesis of 1-((5-oxo-4,5-Dihydro-1,3,4-oxadiazol-2-yl)methyl)-1H-1,2,4-triazole-3-carboxylic Acid-Application of Antibacterial activity Manuri Brahmayya, Dasi Samsonu, Alaparthi Venkateswara Rao, Nelli Srinivasa Rao, Battula Venkateswara Rao, Huan-Jung Fan Proceedings of the National Academy of Sciences India Section A Physical Sciences, 2019 Synthesis of 1-((5-oxo-4,5-dihydro-1,3,4-oxadiazol-2-yl) methyl)-1H-1,2,4-triazole-3-carboxylic acid was accomplished starting with the commercially available 1,2,4-triazole-3-carboxylic acid. The conversion of hydrazide and carbon dioxide to 1,3,4-oxadiazole has been explained. Antibacterial activity also studied for some synthesized compounds.
Bone regeneration using adipose-derived stem cells in injectable thermo-gelling hydrogel scaffold containing platelet-rich plasma and biphasic calcium phosphate Han Liao, Ming-Jin Tsai, Manuri Brahmayya, Jyh-Ping Chen International Journal of Molecular Sciences, 2018 For bone regeneration, a biocompatible thermo-gelling hydrogel, hyaluronic acid-g-chitosan-g-poly(N-isopropylacrylamide) (HA-CPN) was used as a three-dimensional organic gel matrix for entrapping rabbit adipose-derived stem cells (rASCs). Biphasic calcium phosphate (BCP) ceramic microparticles were embedded within the gel matrix as a mineralized bone matrix, which was further fortified with platelet-rich plasma (PRP) with osteo-inductive properties. In vitro culture of rASCs in HA-CPN and HA-CPN/PRP/BCP was compared for cell proliferation and osteogenic differentiation. Overall, HA-CPN/PRP/BCP was a better injectable cell carrier for osteogenesis of rASCs with increased cell proliferation rate and alkaline phosphatase activity, enhanced calcium deposition and mineralization of extracellular matrix, and up-regulated expression of genetic markers of osteogenesis. By implanting HA-CPN/PRP/BCP/rASCs constructs in rabbit critical size calvarial bone defects, new bone formation at the defect site was successfully demonstrated from computed tomography, and histological and immunohistochemical analysis. Taken together, by combining PRP and BCP as the osteo-inductive and osteo-conductive factor with HA-CPN, we successfully demonstrated the thermo-gelling composite hydrogel scaffold could promote the osteogenesis of rASCs for bone tissue engineering applications.
