Mrs. Bhuvaneswari C has more than 17+ years of teaching, including 5 years of research experience in the field of Anatomy. I did my PhD in Medical Anatomy from SRM Medical college hospital and research institute, Chennai, MSc in Human Anatomy at Mudaliar PG institute of basic medical science (IBMS), Taramani, University Madras in the 2002. My field of interest includes Osteoporosis and herbal drug discovery. I worked in grades of Tutor, Assistant professor and Associate professor in Department of Anatomy, SRM medical college hospital and research centre, Potheri, Kanchipuram dist. and working at Panimalar Medical College and reserach institute. Currently I'm fascinated towards research on the effect of medicinal plants on osteoporosis, which made me to take over this research area.

IMMUNO HISTOLOGICAL EVIDENCE OF ORMOCARPUM SENNOIDES EXTRACT IN THE EXPRESSION OF BAX AND BCL2 IN WISTER RATS WITH GLUCOCORTICOID INDUCED OSTEOPOROSIS

Objective: Evaluate the effect of Ormocarpum sennoides through immunohistological evidence in the expression of Bax and Bcl2 in glucocorticoid induced Osteoporosis in Wister rats Method: Rats were divided into five groups (Control, MPA(Osteoporosis), Alendronate (2mg/kg), 100mg (EOS) and 200mg (EOS). The treatment were conducted for 45 days the Bax and Bcl2 expression were evaluated in osteoclast of distal femur diaphysis.

    Knee osteoarthritis (KOA) is a leading cause of pain and functional disability worldwide. Radiographic severity grading using the Kellgren–Lawrence (KL) scale is widely adopted in clinical practice but remains subjective and susceptible to inter- and intra-observer variability, particularly when distinguishing adjacent grades. This study aims to develop an accurate, interpretable, and anatomically guided framework for automated KOA severity grading from plain knee radiographs. A novel Radiomic Feature Fusion and Joint Space Segmentation (RFF-JSS) framework is proposed for automated KOA severity assessment. The pipeline includes (i) standardized preprocessing of knee radiographs, (ii) anatomically informed tibiofemoral joint space segmentation using a U-Net model, (iii) IBSI-compliant multiscale radiomic feature extraction from medial and lateral compartments, (iv) PCA-based radiomic feature fusion for dimensionality reduction and redundancy suppression, and (v) ordinal-aware KOA severity classification using a Random Forest classifier. The framework was evaluated on 5,820 knee radiographs from two large public cohorts—the Osteoarthritis Initiative (OAI) and the Multicenter Osteoarthritis Study (MOST)—with stratified training, validation, and testing splits. The proposed RFF-JSS framework achieved an accuracy of 94.87%, precision of 95.12%, sensitivity of 94.68%, specificity of 96.10%, F1-score of 94.89%, ROC-AUC of 0.972, and a Quadratic Weighted Kappa (QWK) of 0.947. The method consistently outperformed state-of-the-art approaches, including ResNet-SVM, UNet-Radiomics, Deep-KOA, JSN-Net, RF-RFE, and CNN-KLNet. Ablation studies confirmed the critical contributions of joint space segmentation, compartment-wise analysis, feature standardization, and PCA-based radiomic fusion in improving classification robustness and reducing misclassification between adjacent KL grades. The RFF-JSS framework provides a robust, interpretable, and anatomically grounded solution for automated KOA severity grading from plain radiographs. By synergistically combining joint space segmentation with radiomic feature fusion and ordinal-aware classification, the proposed approach bridges the gap between high predictive performance and clinical interpretability, demonstrating strong potential for large-scale screening and clinical decision support in knee osteoarthritis assessment.

    Introduction: Glucocorticoid therapy is the most common cause for secondary osteoporosis leading to a resurgence of interest in traditional medicine that could have bone sparing effects by naturally occurring bioactive molecules. Therefore, this study was done to evaluate the effect of Ormocarpum sennoides DC through immunohistological evidence in the expression of Bax and Bcl2 in glucocorticoid-induced Osteoporosis in Wister albino rats. Materials and Methods: Rats were divided into five groups (Control, MPA (Osteoporosis), Alendronate (2 mg/kg), 100 mg (EOS), and 200 mg (EOS). The treatment was conducted for 45 days the Bax and Bcl2 expressions were evaluated in osteoclast of distal femur diaphysis. Results: When compared the MPA to control group (P < 0.05) Bax increased and Bcl2 reduced, when compared with MPA (P < 0.05) Alendronate, EOS 100 mg and EOS 200 mg increased the Bcl2 expressions, Alendronate, EOS 100 mg, and EOS 200 mg reduced the Bax expression. Discussion and Conclusion: EOS 100 mg and EOS 200 mg increased Bcl2 expression and reduced the Bax expression Ormocarpum sennoides prevents glucocorticoid-induced Apoptosis of preosteoblast; therefore, Bcl2 may be an important regulator of bone growth thus reporting antiapoptotic property of Ormocarpum sennoides DC.
