QbD-guided development of a simvastatin-loaded transethosomal gel for enhanced transdermal delivery: optimization, characterization, and in vivo antihyperlipidemic evaluation Umashri A. Dandagi, Sathgowda Patil Drug Development and Industrial Pharmacy, 2026 BACKGROUND: Simvastatin, a lipid-lowering drug, has low oral bioavailability due to poor solubility and extensive first-pass metabolism. Transdermal delivery may overcome these limitations. OBJECTIVES: The present study aimed to develop, optimize, and evaluate a simvastatin-loaded transethosomal gel for enhanced dermal permeation and improved antihyperlipidemic efficacy. METHODS: Critical quality attributes, including particle size (PS), entrapment efficiency (%EE), and polydispersity index (PDI), were established through a Quality Target Product Profile. Transethosomes were optimized using a Face-Centered Central Composite Design (FCCCD) by varying Soya phosphatidylcholine and ethanol concentrations. The optimized formulation was characterized using dynamic light scattering, scanning electron microscope, and transmission electron microscope. It was incorporated into a 2% w/v carbopol gel and evaluated for pH, viscosity, and spreadability. In vitro diffusion, ex vivo skin permeation, and skin irritation studies were performed. Antihyperlipidemic efficacy was assessed in high-fat diet-induced hyperlipidemic rats, comparing oral simvastatin (10 mg/kg/day) and transethosomal gel (1 g containing 10 mg simvastatin). RESULTS: < 0.05). CONCLUSION: The simvastatin-loaded transethosomal gel demonstrated superior dermal delivery, efficacy, and safety, supporting its potential as a nanoenabled transdermal alternative for hyperlipidemia management.
Supersaturable self-nanoemulsifying orally disintegrating tablets: a novel approach for improved delivery of cinacalcet hydrochloride Vinayak Mastiholimath, Himanshi Koushik, Nisha Shirkoli, Panchaxari M. Dandagi, Atith Muppayyanamath, Rahul Koli Journal of Dispersion Science and Technology, 2025 Cinacalcet hydrochloride (CHCl) has poor solubility and limited permeability across biological membranes, affecting its bioavailability. This study focuses on developing orally disintegrating tablets (ODTs) using supersaturable self-nano emulsifying drug delivery systems (SNEDDS) to enhance CHCl dissolution and absorption. Using the solvent-shift method, CHCl-loaded SNEDDS were formulated and optimized via Scheffé’s mixture design, incorporating Capmul MCM C8 (oil), Kolliphor EL (surfactant), and Span 60 (co-surfactant). Soluplus was used as a precipitation inhibitor polymer. Optimization was based on solubility (86.45 ± 0.19 mg/mL), droplet size (162.37 ± 0.15 nm), percent transmittance (96.3 ± 0.22%), precipitation (12.68 ± 0.31%), and emulsification time (14 ± 0.12 s). The SNEDDS mixture was further solidified using an adsorbent blend of Micel 102 and Micel 112 (2:1 ratio), demonstrating enhanced adsorption efficiency. ODT incorporating Prosolv ODT G2 were prepared via direct compression, ensuring rapid disintegration and wetting time. In vitro studies showed 96.4 ± 0.71% (SD, n = 3) drug release within 30 min. Short-term stability studies indicated no significant degradation over time. The optimized CHCl-loaded SNEDDS-based fast-dissolving tablets significantly improve solubility, dissolution rate, and overall drug stability, presenting a promising strategy to enhance CHCl bioavailability.
A design of expert-based development and optimization of voriconazole-loaded aspasomal gel for topical delivery Shubham Bajirao Patil, Panchakshari Dandagi, Rubeen Dadakalandar Nadaf, Sujay Hulyalkar Journal of Applied Pharmaceutical Science, 2024 The research aimed to develop a voriconazole (VRC)-loaded aspasomal gel for topical drug delivery, to minimize the dosing frequency and associated side effects. Aspasomal formulations were prepared using the thin film hydration method and their vesicle size (VS) and entrapment efficiency (%EE) were assessed. Optimization was carried out using a 32-factorial design by design expert software. The optimized batch underwent in vitro anti-fungal activity testing and morphological analysis using high-resolution transmission electron microscopy. This optimized batch was then incorporated into a gel, and various parameters such as drug content, pH, viscosity, spreadability, in vitro diffusion, ex vivo permeation, and in vivo skin irritancy were evaluated. The optimized aspasomal formulation displayed a VS of 171.36 ± 2.3 nm and an %EE of 80.77% ± 1.75%. In vitro drug diffusion and ex vivo permeation studies demonstrated significant release rates of 94.09% and 48.87% after 24 and 12 hours, respectively. The optimized formulation exhibited better in vitro anti-fungal activity against Candida albicans compared to a commercially available formulation. Skin irritancy tests on rat skin showed no signs of erythema or edema, confirming non-irritation. Long-term stability analysis confirmed the formulation’s stability under cold conditions. In conclusion, VRC-loaded aspasomes present a promising alternative for treating skin fungal infections, offering controlled drug release and minimizing adverse effects, ultimately enhancing patient outcomes.
