PHARMACOLOGICAL ACTIVITY OF 2-(DIETHYLAMINO)ETHYL-4-[(11-OXOINDENE[2,1-B] QUINOXALINE-6-CARBONYL)AMINO]BENZOATE DURING TRANSDERMAL APPLICATION L. V. Eberle, K. D. Sazonov, Ya. V. Rozhkovskyi, S. I. Bohatu, B. V. Prystupa Odesa Medical Journal, 2025 In medical practice, pain syndrome and inflammation are common clinical manifestations that significantly reduce the quality of life of patients. A promising direction of modern pharmacotherapy is the development and search for local dosage forms with analgesic and anti- inflammatory effects. The aim of this work was to study the analgesic and anti-inflammatory activity of a new compound – 2-(diethylamino)ethyl-4- [(11-oxoindeno[2,1-b]quinoxaline-6-carbonyl)amino]benzoate in the composition of ointments of different concentrations. The study was conducted on 50 rats and 60 mice, in accordance with the requirements of GLP and ethical standards. The study of analgesic activity was carried out on models of thermal (“hot water”) and chemical (allyl isothiocyanate test) pain. The assessment of anti-inflammatory activity was carried out on models of exudative inflammation induced by trypsin and zymosan. Research results. According to the results of the study, in the “hot water” test, 5% ointment based on 2-(diethylamino)ethyl-4- [(11-oxoindeno[2,1-b]quinoxaline-6-carbonyl)amino]benzoate provided a significant reduction in pain sensitivity by 63.8%, and in the allyl isothiocyanate test — 88 to 66.5% compared to the control group, which indicates a pronounced analgesic activity of the studied compound. In the model of inflammation induced by zymosan, a moderate anti-exudative effect was established, which was manifested by a decrease in the inflammatory reaction by 27.4%, while in the trypsin model no significant effect was recorded. Thus, it was established that the studied compound is characterized by pronounced local analgesic activity and limited anti-inflammatory properties, which depend on the type of model. The obtained results indicate the feasibility of further preclinical studies of the drug as a potential local analgesic with a combined mechanism of action.

STUDY OF THE WOUND-HEALINGAND REPARATIVE EFFECT OFAN OINTMENT BASED ON THE EXTRACT OF TRIBULUS TERRESTRIS L. ON MODELS OF ASEPTIC FLAT AND LINEAR CUT WOUNDS IN RATS Ya. V. Rozhkovskyi, S. I. Bohatu, B. V. Prystupa, P. B. Antonenko Odesa Medical Journal, 2024 One of the promising ways to improve wound healing is the creation and wider introduction into clinical practice of natural ointments with pronounced anti-inflammatory and reparative properties, which are able to provide a complex therapeutic effect on treating skin wounds. The aim of the study is to determine the wound-healing and reparative effect of ointment with Tribulus terrestris L. extract in experimental wound damage to the skin in rats. Materials and methods. In experiments on 80 non-linear white rats of both sexes, it was analyzed the comparative wound-healing and reparative effect of an ointment based on the extract of Tribulus terrestris L., which was created at the Department of General and Clinical Pharmacology and Pharmacognosy of the Odesa National Medical University, and the comparison drug – methyluracil ointment. The comparative wound-healing effect of ointments was studied on the model of an aseptic planar skin wound in rats and evaluated by the planimetric method. The reparative effect of ointments was studied on a model of an aseptic linear incised skin wound and evaluated by the strength of the scar formed during wound healing in rats using the wound tensiometry method. Treatment was carried out by daily one-time application of ointments to the wound surface using an open method. Results. The property of an ointment based on the extract of Tribulus terrestris L. to shorten the healing time of an aseptic planar skin wound in rats by 1.5–2.0 times was established. In addition, it was shown that in terms of the expressiveness of the wound-healing effect in all terms of the experiment, the ointment under study is not inferior to the comparison drug – methyluracil ointment. In the model of aseptic linear incised skin wound in rats, ointment based on Tribulus terrestris L. extract is superior to methyluracil ointment in terms of reparative activity. The established therapeutic effect of the ointment based on the extract of Tribulus terrestris L. opens up prospects for creating a new ointment based on it for the effective treatment of wound processes of the skin of various genesis.
DOSE-DEPENDENT EFFECT OF HEXADECYL ESTER OF IBUPROFEN ON THE MODEL OF FORMALIN-INDUCED INFLAMMATION B. V. Prystupa, S. I. Bogatu, V. Y. Kresyun, I. A. Boiko, L. M. Unhurian Odesa Medical Journal, 2024 Introduction. Inflammation is a protective reaction of the organism to the damaging agent. Ibuprofen has anti-inflammatory, antipyretic, and anti-edema effect. But ibuprofen has also side effects. The main goal was to estimate the “dose-effect” dependence of the hexadecyl ester of ibuprofen on the formalin model of inflammation. Materials and methods. An acute inflammatory reaction was caused by the subplantar injection of 0.1 ml of 2% formalin into the hind limb of experimental rats. After the development of the inflammatory process, rats were treated by transdermal administration of the hexadecyl ester of ibuprofen as an ointment containing the ester in various concentrations – 5%, 2.5%, 1% and 0.5%. The dynamics of change in the inflammatory process determined by measuring the width and volume of the affected limbs. Results. The hexadecyl ester of ibuprofen reduces the width and volume area of the inflammation in animals and has anti-inflammatory activity at the concentrations 0.5–5%. Conclusion. The hexadecyl ester of ibuprofen demonstrated the high anti-inflammatory activity. The anti-inflammatory activity was not dependent from the concentration. It is advisable to use the 0.5% ointment of ibuprofen ester, which is almost equal in their efficiency to the 5% ointment.
