A Novel Perspective in Drug Design and Drug Discovery (4D) With Computer-Aided Drug Design (CADD): A Review Sharad R Manapure, Shruti Kolte, Vipinchandra B Pande, Saket Singh Chandel, Pranjali Khanke, Kanchan Kohale International Journal of Drug Delivery Technology, 2026 Nowadays there are so many types of diseases that are causing to human and animal health. This disease or disorder makes human health very severe, lethal, and life-threatening to survive. Several troubling diseases make the body's condition abnormal or cause a disturbance in the function and structure of the body or organ systems. It states that the person is ill or sick which is characterized by the manifestation or symptoms which may be seen internally or externally. To overcome these manifestations there are some specific substances or molecules are used. These substances are called drugs and can cure various diseases or disorders. A drug compound may be natural or synthetic obtained from a plant, animal sources, or any chemical compound which can treat and prevent diseases. There are some modern methods to produce a drug. To discover a new drug is called a drug discovery which saves time and trial discovery takes a lot of time for selecting, researching, optimization, of a drug. These drugs are beneficial and lifesavers. Several days or years pass or are required to discover a new molecule or to design a single molecule. Drug discovery is the research of new compounds and new moiety. Drug design is a very convenient and time-saving method for drug discovery. Drug design is a type of invention and it is a computational method and it is an important part of drug discovery. There are two techniques of drug design ligand-based and receptor-based drug design. Computer-aided drug design is a computational method in which there is a prediction of the structure and molecules by a mathematical equation. In LBDD QSAR & Pharmacophore model is there and in RBDD Molecular Docking & De Novo Design is present. In QSAR there are of the efficacy of a biologically active compound. It is a mathematical relationship that identifies and quantifies the molecule. These methods can be applied and then virtual screening. Then the identification of the molecule binds to the binding site or receptor and gives its action.
A Novel Perspective on Pharmaceutical Research: Mucoadhesive Patches Sharad R Manapure, Vipinchandra B Pande, Pranjali Khanke, T. Venkatachalam, Saket Singh Chandel International Journal of Drug Delivery Technology, 2025 Mucosal drug delivery explores the significance of mucoadhesive drug delivery systems in enhancing drug bioavailability and sustained release. It delves into the mechanisms of muco-adhesion, highlighting the role of various polymers. The review also discusses challenges and recent advancements in formulating effective mucoadhesive drug delivery systems, offering insights into their potential applications for targeted and localized drug delivery. It examines the diverse range of polymers employed in mucoadhesive formulations, such as chitosan, alginate, and Carbopol, discussing their specific characteristics and advantages. This addresses the influence of physiological factors, such as pH and mucus composition, on the performance of mucoadhesive systems. Furthermore, light on recent advancements in the field, including the integration of nanotechnology and innovative polymer blends to optimize mucoadhesive properties. It emphasizes the potential of mucoadhesive drug delivery systems in achieving site-specific targeting, minimizing side effects, and improving patient compliance. In addition, the abstract highlights challenges associated with mucoadhesive formulations, including variability in mucosal surfaces and potential toxicity concerns, while proposing strategies to address these issues. A mucosal patch is a drug delivery system designed to adhere to mucosal surfaces, such as those found in the oral cavity, nasal passages, or vaginal area. These patches are formulated with mucoadhesive materials that enable them to stick to the mucosal membrane, allowing for controlled drug release directly at the site of application. The mucosal patch offers advantages like enhanced bioavailability, rapid onset of action, and reduced systemic side effects. These patches can be designed for various therapeutic purposes, including local treatment of mucosal infections, hormone delivery, or management of conditions affecting specific mucosal tissues. In addition to mucoadhesive polymers, mucosal patches may incorporate drug-loaded nanoparticles or microparticles for sustained release.
Comparative Analysis in Hepatoprotective Activity of Crude Extracts of important Medicinal Plants Jitendra Patel, G. Shiva Kumar, Khemkaran Ahirwar, Mukesh K. Gupta, Sandeep K. Singh, Saket S. Chandel, Vijay K. Patel Research Journal of Pharmacy and Technology, 2023 According to the World Health Organization, chronic liver diseases account for approximately 46% of all diseases and 59% of all deaths worldwide. It is critical to look for hepatoprotective crucial findings because they are important for human health. The aim of this study was to compare the hepatoprotective activity of six medicinal plants based on biochemical reduction on the animal study. The increased levels of ALT, AST, ALP, total bilirubin, direct bilirubin, and cholesterol in the blood have been marked to the damaged structural virtue of the liver. When the plant extract was given to rats after they were administered a hepatotoxicant and the level of marker enzymes was discovered to be lower than normal, it meant that the particular dose of the plant extract was considerably supporting the hepatoprotective action. The comparative study of hepatoprotective efficacy of six medicinal plants was determined by a biostatistical analysis of data received from an animal investigation. The Terminalia coriacea and Artocarpus hirsutus at 250 and 500mg/kg respectively revealed that the *P<0.05; **P< 0.01; ***P<0.001; when compared to the CCl4 group. The Cucumis melo revealed P<0.05 and P<0.01 as compared with the liver-damaged control to the drug-treated animal at doses of 100, 250, and 500mg/kg. Buchanania lanzan revealed significant at P<0.05*, 0.01** and 0.001*** at 100 mg/kg, 200mg/kg and 400 mg/kg for aqueous and alcoholic extract both. The Diospyros melanoxylon (Roxb) and Solanum americanum Mill at doses 100, 200, and 400 mg/kg and 200, 400, and 600 mg/kg respectively. The T. coriacea and A. hirsutus revealed a 5%, 1%, and less than 1% chance of false-positive, which is statistically significant at all levels.
Novel Antidiabetic Polyherbal Formulation for Synergistic Therapeutic Effects in Streptozotocin (STZ)-Induced Diabetic Rats Pande V. Bhaskarrao, Chandel S. Singh, Soni . Vishal International Journal of Drug Delivery Technology, 2022 Worldwide demand for new anti-diabetic drugs from plant sources has increased as diabetes mellitus has become a global epidemic. In the era of herbal medicines, polyherbal formulations offer higher therapeutic efficacy than single plants due to synergistic effects. Therefore, the objective was to develop a novel anti-diabetic polyherbal formulation containing mixtures of three plants: Azadirachta indica leaves, Tinospora cordifolia stem and Ocimum sanctum leaves extracts. The eight different plant formulations (F1 to F8) were formulated while F1 to F3 contained a single plant extract. Hyperglycemia was developed in rodents by ingestion of streptozotocin. The experimental animals’ serum sugar level, body weight and lipid profile were determined. In the diabetic rats treated orally with F1 to F8, the blood glucose level decreased significantly compared to the diabetic control group. Similar effects were also observed in the diabetic rats treated with glibenclamide. In addition, F1 to F8 controlled lipid level, namely total cholesterol (CHL), triglycerides (TGL), low-density lipoprotein (LDL), and high-density lipoprotein (HDL) levels in rodents. The findings suggested that F7 showed higher anti-diabetic and antihyperlipidemic efficiency when equated to the other formulations. It was also found that the formulations (F1 to F3) containing a single plant extract exhibited lower therapeutic efficacy than the polyherbal formulations (F4 to F8). The results suggest that the higher therapeutic efficacy of the polyherbal formulation is due to the synergistic effect of the different phytoconstituents in the plant mixtures.
