In-vitro evaluation of chitosan against bacterial pathogens of silkworm, Bombyx mori L. BHUVANESHWAR RAJESH NAIK, N. AMARNATHA, K. C. NARAYANASWAMY, MANJUNATH GOWDA, D. V. NAVEEN Journal of Biological Control, 2025 The present research work was carried out to know the efficacy of the silkworm pupal chitosan against the bacterial pathogens of silkworm Bombyx mori L. under in-vitro conditions. Chitosan was isolated from silkworm pupae and their exuviae through a meticulous three stage extraction process viz., deproteinisation, demineralisation and deacetylation. The extracted chitosan, prepared in various concentrations (i.e., 500-5000 ppm), was rigorously evaluated against the pathogenic bacteria of B. mori (Staphylococcus aureus and Bacillus thuringiensis) under in-vitro conditions. These tests were conducted using nutrient agar plates, with the efficacy of each concentration benchmarked against standard ampicillin and a commercial chitosan preparation by assessing their inhibitory effect through measuring the inhibition zones around treated discs. The antibacterial activity increased with chitosan concentration, with the highest zone of inhibition observed at 5000 ppm (1.191 cm for S. aureus and 1.100 cm for B. thuringiensis), which was on par with the treatment ampicillin and commercial chitosan at 1000 ppm. Concentrations ≥2000 ppm showed antibacterial efficacy comparable to standards at 1000 ppm. The antimicrobial action is attributed to the interaction between positively charged amino groups of chitosan and bacterial teichoic acids, resulting in cell wall rupture and death of bacteria. The in-vitro study reveals that, silkworm pupal chitosan exerts potent antibacterial activity against S. aureus and B. thuringiensis, particularly at ≥4,500 ppm. This valorises a sericultural by-product, offering a sustainable alternative to synthetic bactericides. The findings hold promise for enhancing silkworm health management and optimising sericulture productivity.
