Bevza O.V. in 1992 graduated from the Taras Shevchenko National University of Kyiv (Faculty of Biology, Department of Biochemistry) with a degree in biologist - biochemist, teacher of biology and chemistry. Since 1987, he has been working at the O.V. Palladin Institute of Biochemistry of the National Academy of Sciences of Ukraine in the following positions: laboratory assistant (1987-1990), engineer (1990-1997), acting junior research fellow (1997-1998), research fellow (since 1998), senior research fellow (since 2021). He defended his PhD thesis in 1998. Bevza O.V. is a specialist in the field of biochemistry. He is actively involved in the methods of preparative biochemistry, chemical and biochemical kinetics, biochemical membrane science, spectrophotometry, spectrofluorimetry, laser correlation spectroscopy, enzymology, and statistical analysis. Thiacalix[4]arene С-1193 – a promising inhibitor of the sodium pump in the uterine smooth muscle cells , O. V. Maliuk, T. O. Veklich, , O. V. Tsymbalyuk, , O. V. Bevza, , S. O. Cherenok, , A. I. Kosterin, and Ukrainian Biochemical Journal, 2025 Thiacalix[4]arene C-1193 (25,27-dibutoxythiacalix[4]arene-bis-hydroxymethylphosphonic acid) was shown to inhibit the activity of Na+,K+-ATPase with a high efficiency (І0.5 = 42.1 ± 0.6 nM) with no effect on the activity of Mg2+-ATPase, Са2+-ATPase and Са2+,Mg2+-ATPase in the plasma membrane fraction of rat uterine smooth muscle cells. The kinetic regularities of the C-1193 inhibitory effect on Na+,K+-ATPase activity were investigated. It was demonstrated that C-1193 increased the enzyme activation constant by Na+ but not by K+ ions. The contractile activity of the rat uterine horns was investigated by tenzometric methods with the use of longitudinal uterine smooth muscle strips with intact endometrium. С-1193 induced a considerable increase in the amplitude of the acetylcholine-induced contractions as well as the maximal velocity of the contraction and relaxation phases. No effect of С-1193 on contractive activity induced by the selective agonist of М3-cholinoreceptors cevimeline was observed. The results of computer simulation showed that С-1193inhibitory effect must be related to the cooperative action of methylene bisphosphonate fragments on the upper rim of the calixarene platform, and the linker sulfur atoms of calixarene “cup” on the Na+,K+-ATPase macrostructure. Thiacalix[4]arenes Remove the Inhibitory Effects of Zn Cations on the Myosin ATPase Activity Raisa Labyntsevа, Viktoriia Yavorovska, Olexander Bevza, Andriy Drapaylo, Vitaly Kalchenko, Sergiy Kosterin Nanoscale Research Letters, 2018 Numerous female reproductive abnormalities are caused by uterine smooth muscle (myometrium) disorders. Heavy metals have an adverse effect on the contractility of the uterine smooth muscle. Although zinc is an essential biogenic element for most of the organisms, high doses of this element are toxic. The study of 0.5−5 mM Zn2+ effect on myosin S1 ATPase activity from the uterus found that 5 mM Zn2+ cations have the most pronounced inhibitory effect. The calculation of the kinetic parameters (Km and Vmax, ATP) revealed that the apparent maximum velocity of the hydrolysis ATP catalyzed by myosin in the presence of 5 mM Zn2+ decreased by 1.6 times. The value of Кm for ATP hydrolysis by myosin S1 in the presence of Zn2+ does not change statistically, although it tends to decrease. It was determined that uterine myosin S1 ATPase activity does not depend on the concentration of Mg2+ in the presence of 5 mM Zn2+. Also, it was demonstrated that tetrahydroxythiacalix[4]arene-tetrasulfosphonate (C-798) and tetrahydroxythiacalix[4]arene-tetraphosphonate (C-800) restored myosin S1 ATPase activity to the control level in the presence of 5 mM Zn2+. One of the most probable mechanisms of restoring the action of these thiacalix[4]arenes protective effect is based on its ability to chelate heavy metal cations from the incubation medium. The molecular docking of C-798 and C-800 into the myosin S1 region showed that these thiacalix[4]arenes could interact with Zn cation bond by myosin amino acid residues near the ATPase active site. Therefore, thiacalix[4]arenes may weaken the interaction between this cation and myosin S1. It was speculated that the obtained results could be used for further research with the aim of using this thiacalix[4]arenes as pharmacological compounds in the case of poisoning with high concentrations of zinc.

Kosterin, and Ukrainian Biochemical Journal, 2016 Numerous female reproductive abnormalities are consequences of disorders in uterus smooth muscle (myometrium) contractile function. In this work, we described activators of ATPase, which could be used for development of effective treatments for correcting this dysfunction. Myosin ATPase localized in the catalytic domain of myosin subfragment-1 transforms a chemical energy deposited in macroergic bonds of ATP into mechanical movement. It was shown that сalix[4]arene C-90 and its structural analogs functionalized at the upper rim of macrocycle with four or at least two N-phenylsulfonуltrifluoroacetamidine groups, are able to activate ATP hydrolysis catalyzed by myometrium myosin subfragment-1. It was shown with the method of computer modeling that N-phenylsulfonуltrifluoroacetamidine groups of calix[4]arene C-90 interact with responsible for binding, coordination and the hydrolysis of ATP amino acid residues of myosin subfragment-1. The results can be used for further research aimed at using calix[4]arene C-90 and its analogs as pharmacological compounds that can effectively normalize myometrium contractile hypofunction.
Protective effect of tiacalix[4]arene-tetrasulphonate on heavy metal inhibition of myometrium myosin subfragment-1 ATP-hydrolase activity and R. D. Labyntsevа Ukrainian Biochemical Journal, 2014 Heavy metals have a negative effect on the contractility of uterine smooth muscles (myometrium), these effects can lead to various pathologies of a women reproductive system. To overcome these effects the methods for correcting the myometrium contractile activity are to be developed. Catalyzed by myosin ATPase ATP hydrolysis is the most important reaction in the molecular mechanism of myometrium contraction. We have found an inhibitory effect of 0.03-0.3 mM Ni2+, Pb2+ and Cd2+ on enzymatic hydrolysis of ATP by myosin subfragment-1 obtained from swine uterine smooth muscles. We have demonstrated that 100 μM thiacalix[4]arene-tetrasulphonate (C-798) recovered to the control level of ATPase activity of myosin subfragment-1 in the presence of heavy metal cations. One of the most probable mechanisms of C-798 corrective activity is based on its ability to chelate heavy metals, thus cations Pb, Cd and Ni can be removed from the incubation medium. Computer simulation has demonstrated that the protective effect of C-798 may also be the result of weakening the interaction of heavy metal ions with amino acid residues of the myosin molecule near the active site of ATP hydrolase. The obtained results can be used for further research aimed at assessing the prospects of thiacalix[4]arene-tetrasulfonate as pharmacological compounds.
