- Targeting glioblastoma with HDAC inhibitors: insights into hydroxamic acid-based therapeutic strategies

Epigenetic modifications play crucial roles in glioblastoma growth and aggressiveness, with key regulators including histone deacetylases (HDACs), histone acetyltransferases (HATs), and methyltransferases. Targeting these epigenetic alterations has emerged as a promising therapeutic strategy, utilizing DNA methyltransferase (DNMT) inhibitors, HDAC inhibitors (HDACis), and miRNA-based therapies. HDACis, whose effect on p53, p21, Bax, and Bcl-2, have gained significant interest because of their ability to restore the expression of tumor suppressor genes, thereby inducing apoptosis and overcoming therapeutic resistance. Our study demonstrated that a novel hydroxamic acid analogue, compound 3B, effectively inhibited glioma cell (C6) proliferation and exhibited potent anticancer activity. Compound 3B induced G2/M phase cell cycle arrest, increased apoptotic cell populations, and significantly reduced colony formating efficiency. Confocal imaging revealed nuclear condensation and elevated reactive oxygen species (ROS) levels, indicating oxidative stress. Western blot analysis confirmed that HDAC inhibition increased AcH3K9 protein levels. Further, studies in in vivo xenograft model and allograft C6 Wistar rat model revealed strong antitumour activity, suggesting that compound 3B is a promising therapeutic candidate for glioblastoma treatment. - In silico and in vitro evaluation of risedronate as a DKK1 antagonist: Implications for neuroprotection through DKK1–LRP6 modulation

Dickkopf-1 (DKK1) mediated dysregulation of the Wnt/β-catenin signaling pathway contributes to synaptic loss in Alzheimer's disease (AD). Risedronate, a third-generation bisphosphonate, shows promise in pathologies related to the Wnt pathway, although its direct interaction with the DKK1-LRP6 complex has yet to be investigated. This study aimed to characterize risedronate as a novel small-molecule DKK1 antagonist and evaluate its capacity to restore canonical Wnt signaling and provide neuroprotection. In silico studies were performed using the Schrödinger Maestro (v2018-3) platform, utilizing molecular docking, molecular simualtion, SiteMap and WaterMap analysis to define the thermodynamic hydration energetics of the DKK1-LRP6 interface. In vitro validation was conducted using LRP6-overexpressing HEK-293T cells, with Wnt modulation quantified via immunofluorescence of membrane-associated DKK1 and β-catenin stabilization assays and neuroprotective assay in presence of glutamate in SHSY5Y cells. In silico analysis identified a high-affinity binding site on the DKK1 C-terminal domain (Dscore: 0.998). WaterMap analysis and simulation study revealed that risedronate binding is driven by the displacement of high-energy "unfavorable" water molecules (ΔG > 0) at the Arg203/Lys222 residue cluster. In HEK-293LRP6 cells, risedronate (IC 50 : 65.94 μM) significantly attenuated DKK1-mediated LRP6 sequestration (p < 0.0001), acting as a molecular "plug" to restore β-catenin signaling. This restoration successfully suggests a potential modulation of downstream of Wnt pathway restoration. Risedronate demonstrates potential inhibitory effects on DKK1 in in vitro and in silico models. By bridging bone pharmacology and neurobiology, this study provides a mechanistically grounded repurposing strategy to counteract Wnt signaling failure in neurodegenerative disorders. - Targeting P2X7 receptor/NLRP1 inflammasome axis and gut dysbiosis: A mechanistic review of pyroptosis in metabolic inflammation

BACKGROUND: Obesity and type 2 diabetes (T2D) are chronic metabolic disorders characterized by persistent low-grade systemic inflammation. While the NLRP3 inflammasome is well-documented, the NLRP1 inflammasome has recently emerged as a proximal molecular ""tripwre" that initiates pyroptotic cell death and cytokine maturation, driving metabolic decline. MECHANISMS: Hyperglycemia and lipotoxicity induce cellular stress, leading to the release of extracellular ATP (eATP) through Pannexin-1 channels. Simultaneously, dysbiosis-induced "leaky gut" facilitates the systemic translocation of lipopolysaccharides (LPS). These DAMPs and PAMPs activate the P2X7 receptor (P2X7R) and Toll-like receptor (TLR) signaling pathways, respectively, triggering the NLRP1 inflammasome. Once activated via functional degradation, the human NLRP1 C-terminal fragment recruits caspase-1, either directly via its CARD domain or through the ASC adaptor, to cleave gasdermin D (GSDMD) and pro-inflammatory cytokines (IL-1β and IL-18). This process executes inflammatory pyroptosis in adipocytes and pancreatic β-cells. Crucially, NLRP1 also serves as an intrinsic negative regulator of the Th17/STAT3 axis; its dysregulation leads to uncontrolled IL-17 production and the activation of JNK/NF-κB pathways. This signaling cascade inhibits the IRS-1/PI3K/AKT axis and impairs insulin secretion by downregulating GLUT2, PDX-1, and GCK. CONSEQUENCES AND TREATMENTS: Chronic NLRP1 hyperactivation fosters a self-perpetuating "vicious cycle" of tissue damage, insulin resistance, and β-cell attrition, leading to macrovascular and microvascular complications. To disrupt this cascade, a multi-faceted therapeutic approach is required. Restoring the intestinal barrier through probiotics and short-chain fatty acids (SCFAs) can limit the circulation of inflammatory primers. Concurrently, targeting the P2X7R/NLRP1 axis with specific antagonists and inflammasome inhibitors provides a pragmatic strategy to reduce systemic inflammation. Myocardial infarction (MI) and stroke are examples of ischaemic diseases that continue to pose serious health risks since few specific treatments address the underlying molecular causes of these conditions, which include pathological thrombosis and mitochondrial dysfunction. In order to close this gap, we used an integrated in-silico method to find strong inhibitors for Protease-Activated Receptor-1 (PAR-1) and Cyclophilin D (CypD) from the structurally diverse CMNPD marine natural product library. To account for protein flexibility, our method combines High Throughput Virtual Screening (HTVS), Standard Precision (SP), and Extra Precision (XP) docking with meticulous Induced Fit Docking (IFD). After undergoing 500 ns Molecular Dynamics (MD) simulations, the best candidate compounds showed remarkable structural stability with RMSD values between 0.6 and 1.7 Å. Strong binding affinities were found by thermal MM-GBSA calculations, with CMNPD29955 showing a ΔG of –63.146 kcal/mol towards CypD and CMNPD3572 and CMNPD22037 showing ΔG values of –96.24 and –98.092 kcal/mol against PAR-1, respectively. Lead molecules of each target were found to have important hydrophobic and hydrogen bonding interactions with crucial active site residues in MD modelling. The dynamic stability and energetic favorability of these chemicals were further confirmed by complementary techniques, including WaterMap, Principal Component Analysis (PCA), and Free Energy Landscape (FEL). In addition to CMNPD3572 and CMNPD22037 as potential PAR-1 antagonists, this work emphasises CMNPD29955 as a new lead inhibitor that targets mitochondrial dysfunction via CypD. To transform computational insights into effective and complementary therapeutic alternatives, lead optimisation, experimental validation, and preclinical assessment of these special marine-derived molecules will be crucial in the future. • By focusing on thrombosis (Protease-Activated Receptor-1 (PAR-1)) and mitochondrial dysfunction (Cyclophilin D (CypD)), an integrated multi-stage in-silico workflow was used to find new treatments for ischemic diseases. • Lead compounds demonstrated excellent dynamic stability during 500 ns molecular dynamics simulations, with RMSD values consistently between 0.6 and 1.7 Å. • MM-GBSA binding free energy calculations revealed strong affinities, including CMNPD3572 and CMNPD22037 for PAR-1 (ΔG ≈ -96 to -98 kcal/mol) and CMNPD29955 for CypD (ΔG ≈ −63 kcal/mol). • Further advanced analyses, such as WaterMap, PCA and Free Energy Landscape studies, confirmed favorable hydration thermodynamics along with stable conformational dynamics and energetically preferred binding states. • CMNPD29955 emerged as a novel marine-derived CypD inhibitor. Heterocyclic scaffolds are central to drug discovery due to their diverse pharmacological properties. In this work, novel pyridine and 1,3,4-oxadiazole linked 1,3-thiazole hybrids were designed and synthesized as potential anticancer agents. Their structures were confirmed by various spectroscopic methods, and the compounds were evaluated for cytotoxicity against the MCF-7 breast cancer cell line. Among the series, compound 7a (IC₅₀ = 28.51 ± 1.70 µM) displayed the most potent activity, approaching that of cisplatin (23.44 ± 2.23 µM). Substituent effects revealed that a para-methyl group enhanced activity, whereas electron-withdrawing groups reduced potency. Computational studies (docking, MD simulations, MMGBSA, DFT, and drug-likeness predictions) further supported the experimental findings and highlighted favorable binding interactions. A novel series of twelve quinazoline-based oxadiazole hybrids was synthesized via a concise four-step synthetic route and comprehensively characterized by FTIR, 1 H NMR, 13 C NMR, and high-resolution mass spectrometry. Single crystal-XRD results confirmed that the para-fluoro-substituted derivative adopts a monoclinic crystal system and is assigned to the P2₁/c space group. The molecular structure featured a nearly coplanar arrangement of the oxadiazole and quinazoline rings, with a dihedral angle of 1.5°, while the fluorophenyl–oxadiazole linkage displayed a dihedral angle of 12.8°, suggesting slight torsional deviation. Hirshfeld surface analysis indicated that intermolecular contacts such as H···F (26.3%), H···N (8.7%), H···O (7.3%), and π···π stacking interactions play a key role in molecular packing and lattice stability. Molecular docking studies were conducted to evaluate the interaction of the synthesized hybrids with histone deacetylase 7 (HDAC7), an epigenetic target implicated in oncogenesis. The docking analysis revealed favorable binding within the HDAC7 active site. Furthermore, a 100 ns molecular dynamics simulation revealed that the HDAC7–ligand complex maintained structural integrity, exhibited minimal RMSD deviations, and preserved key interactions throughout the simulation. ADME predictions using the QikProp module suggested favorable pharmacokinetic properties and drug-likeness. Collectively, the results offer comprehensive structural and computational insights into quinazoline–oxadiazole hybrids, providing a foundation for future biological evaluation. - Quantum Dot Localized Surface Plasmon Resonance Biomedical Applications

Cerebral ischemia is a major neurological condition that contributes significantly to global morbidity and mortality. It occurs due to reduced blood supply to the brain, leading to neuronal injury and death. The pathophysiology of cerebral ischemia is complex and involves oxidative stress, neuroinflammation, excitotoxicity, mitochondrial dysfunction, and apoptosis. Despite advances in medical management, effective neuroprotective therapies remain limited, creating a need to explore alternative approaches. Mucuna pruriens is a medicinal plant widely used in traditional medicine, known for its rich content of bioactive compounds, including levodopa, flavonoids, alkaloids, and phenolic antioxidants. These compounds are associated with antioxidant, anti-inflammatory, and neuroprotective properties. This review summarizes the available evidence from animal model studies on the role of M. pruriens in the management of cerebral ischemia. The literature indicates that M. pruriens can reduce oxidative stress by enhancing antioxidant enzyme activities, including superoxide dismutase, catalase, and glutathione peroxidase, and by decreasing lipid peroxidation. Histopathological findings suggest that it helps preserve neuronal structure, particularly in vulnerable brain regions such as the hippocampus. Behavioral studies also demonstrate improvements in motor function, coordination, and cognitive performance following treatment with M. pruriens. These effects are mainly attributed to its ability to scavenge free radicals, modulate neurotransmitter levels, and regulate inflammatory pathways. Although the findings from animal studies are promising, variations in experimental design, dosage, and extraction methods limit the consistency of results. In addition, the lack of standardized formulations and clinical evidence restricts its direct application in human patients. In conclusion, M. pruriens shows potential as a natural neuroprotective agent in cerebral ischemia. However, further well-designed experimental and clinical studies are required to establish its safety, efficacy, and standardized therapeutic use.
