Mahoney, Jones Bernardes Graceli Frontiers in Endocrinology, 2025 Endocrine-disrupting chemicals (EDCs) are a group of substances that can alter normal body functioning by disrupting the various patterns of hormone secretion and action. Some of these substances are used as plasticizers (e.g., bisphenols and phthalates) and agrochemicals (e.g., vinclozolin). EDC exposure can occur by many routes, including oral by contaminated food and water, through the skin, inhalation, and by placental transfer from mother to fetus or mother to infant (via lactation). The increase in EDCs used by the industry has strongly impacted our health. An increasing number of scientific works have reported the effects of EDCs on cancer development, metabolism, heart disease, and fertility. Most recently, studies on EDCs effects on behavior and the developing brain are raising major concerns related to the formation of sex differences and to the increased prevalence of neuropsychiatric disorders. In this review, we highlight the recent findings of the effects of pre-, peri-, and postnatal exposure to the three well-studied EDCs (i.e., bisphenols (BPA, BPS, BPF, and BPAF), phthalates (DBP, BBP, DEHP, and DiPeP), and vinclozolin (VIN)) on developing brain and behavior across generations in experimental animals.

MCT8 is Downregulated by Short Time Iodine Overload in the Thyroid Gland of Rats E. de Souza, G. Dias, R. Cardoso, L. Lima, R. Fortunato, T. Visser, M. Vaisman, A. Ferreira, D. Carvalho Hormone and Metabolic Research, 2015 Wolff-Chaikoff effect is characterized by the blockade of thyroid hormone synthesis and secretion due to iodine overload. However, the regulation of monocarboxylate transporter 8 during Wolff-Chaikoff effect and its possible role in the rapid reduction of T4 secretion by the thyroid gland remains unclear. Patients with monocarboxylate transporter 8 gene loss-of-function mutations and monocarboxylate transporter 8 knockout mice were shown to have decreased serum T4 levels, indicating that monocarboxylate transporter 8 could be involved in the secretion of thyroid hormones from the thyroid gland. Herein, we aimed to evaluate the regulation of monocarboxylate transporter 8 during the Wolff-Chaikoff effect and the escape from iodine overload, besides the importance of iodine organification for this regulation. Monocarboxylate transporter 8 mRNA and protein levels significantly decreased after 1 day of NaI administration to rats, together with decreased serum T4; while no alteration was observed in LAT2 expression. Moreover, both monocarboxylate transporter 8 expression and serum T4 was restored after 6 days of NaI. The inhibition of thyroperoxidase activity by methimazole prevented the inhibitory effect of NaI on thyroid monocarboxylate transporter 8 expression, suggesting that an active thyroperoxidase is necessary for MCT8 downregulation by iodine overload, similarly to other thyroid markers, such as sodium iodide symporter. Therefore, we conclude that thyroid monocarboxylate transporter 8 expression is downregulated during iodine overload and that the normalization of its expression parallels the escape phenomenon. These data suggest a possible role for monocarboxylate transporter 8 in the changes of thyroid hormones secretion during the Wolff-Chaikoff effect and escape.
Diphenyl diselenide modulates gene expression of antioxidant enzymes in the cerebral cortex, hippocampus and striatum of female hypothyroid rats Glaecir Roseni Mundstock Dias, Ronaldo Medeiros Golombieski, Rafael de Lima Portella, Guilherme Pires do Amaral, Félix Antunes Soares, João Batista Teixeira da Rocha, Cristina Wayne Nogueira, Nilda Vargas Barbosa Neuroendocrinology, 2014 <b><i>Introduction:</i></b> Cellular antioxidant signaling can be altered either by thyroid disturbances or by selenium status. <b><i>Aims:</i></b> To investigate whether or not dietary diphenyl diselenide can modify the expression of genes of antioxidant enzymes and endpoint markers of oxidative stress under hypothyroid conditions. <b><i>Methods:</i></b> Female rats were rendered hypothyroid by continuous exposure to methimazole (MTZ; 20 mg/100 ml in the drinking water) for 3 months. Concomitantly, MTZ-treated rats were either fed or not with a diet containing diphenyl diselenide (5 ppm). mRNA levels of antioxidant enzymes and antioxidant/oxidant status were determined in the cerebral cortex, hippocampus and striatum. <b><i>Results:</i></b> Hypothyroidism caused a marked upregulation in mRNA expression of catalase, superoxide dismutase (SOD-1, SOD-3), glutathione peroxidase (GPx-1, GPx-4) and thioredoxin reductase (TrxR-1) in brain structures. SOD-2 was increased in the cortex and striatum, while TrxR-2 increased in the cerebral cortex. The increase in mRNA expression of antioxidant enzymes was positively correlated with the Nrf-2 transcription in the cortex and hippocampus. Hypothyroidism caused oxidative stress, namely an increase in lipid peroxidation and reactive oxygen species levels in the hippocampus and striatum, and a decrease in nonprotein thiols in the cerebral cortex. Diphenyl diselenide was effective in reducing brain oxidative stress and normalizing most of the changes observed in gene expression of antioxidant enzymes. <b><i>Conclusion:</i></b> The present work corroborates and extends that hypothyroidism disrupts antioxidant enzyme gene expression and causes oxidative stress in the brain. Furthermore, diphenyl diselenide may be considered a promising molecule to counteract these effects in a hypothyroidism state.
