- Nrf2/NOX2 Pathway Dysregulation and Oxidative Stress Biomarkers in Gaucher Disease–Associated Parkinsonism: Insights Into a Potential Therapeutic Target

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder, yet its underlying genetic and molecular mechanisms remain incompletely understood. Variants in the GBA gene, encoding the lysosomal enzyme glucocerebrosidase, are not only responsible for Gaucher disease (GD) but also represent a significant genetic risk factor for PD, contributing to lysosomal dysfunction, oxidative stress and autophagy impairment. Among the key regulators of redox homeostasis, the Nrf2/NOX2 signalling axis has emerged as a pivotal pathway in the modulation of neuroinflammation and neurodegeneration. This study aims to explore the pathogenic link between GBA mutations and PD, focusing on the redox imbalance and the role of Nrf2 signalling in an in vivo Gba D409V knock‐in (KI) mouse model, compared to wild‐type (WT) C57BL/6J controls. Animals 8‐weeks old were evaluated over a 3‐month period, with tissue and behavioural assessments conducted at 7, 14, 30, 60 and 90 days. Early timepoints (7 and 14 days) did not reveal significant changes in behavioural performance, expression of PD‐related markers (TH, DAT, α‐synuclein), or oxidative stress indicators, including Nrf2, NOX2, malondialdehyde (MDA) and nitrate/nitrite levels. However, at 30, 60 and especially 90 days, significant alterations emerged, particularly a disrupted Nrf2/NOX2 balance, accompanied by molecular and biochemical signatures of oxidative stress. These findings suggest a time‐dependent progression of oxidative alterations in this GD model and support the role of GBA variants in promoting neurodegenerative processes. The NLRP3 inflammasome, a cytosolic multiprotein complex composed of NLRP3, ASC, and caspase-1, orchestrates the maturation of interleukin-1β (IL-1β) and interleukin-18 (IL-18) and the induction of pyroptosis, acting as a central mediator of innate immunity. Although physiologically protective, aberrant NLRP3 activation has been increasingly implicated in tumorigenesis. In oral squamous cell carcinoma (OSCC), current evidence points to a predominantly pro-tumorigenic role, with elevated NLRP3 expression correlating with tumor progression, lymph node metastasis, advanced pathological stage, and reduced survival. Functional studies demonstrate that genetic silencing or pharmacological inhibition of NLRP3 enhances apoptosis and reduces tumor burden. An additional regulatory layer is provided by microRNAs (miRNAs), which fine-tune NLRP3 expression at the post-transcriptional level. Since the identification of miR-223-3p as the first miRNA to directly target NLRP3, several miRNAs, including miR-22-3p, miR-7-5p, and miR-30e-5p, have been shown to suppress NLRP3 activity in various pathological settings, including oral squamous cell carcinoma, where miR-22-3p downregulates NLRP3, inhibiting its proliferation, migration, and invasion. Therefore, the NLRP3 inflammasome represents a key player in cancer development, and its regulation by miRNAs highlights its importance and clinical potential. This review summarizes mechanistic and clinical knowledge on the biology of NLRP3, highlights its dual role in cancer hallmarks, and discusses the therapeutic promise of targeting the NLRP3-miRNA axis in the management of oral cancer. - Aesculus hippocastanum Extract Exerts Neuroprotective Effects in an MPP+-Induced Parkinson's Disease Model via PPARγ Activation

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterised by the loss of dopaminergic neurons in the substantia nigra. In this study, we investigated the neuroprotective and anti‐inflammatory potential of Aesculus hippocastanum (horse chestnut extract, HCE) in an in vitro model of PD. Human neuroblastoma SH‐SY5Y cells were treated with the neurotoxin 1‐methyl‐4‐phenylpyridinium (MPP + ) (1 mM) and/or in combination with HCE at the concentrations of 15.6, 31.2 and 62.5 μg/mL for 24 h. After 24 h, several analyses have been performed. Treatment with HCE at the concentrations of 31.2 and 62.5 μg/mL significantly improved cell viability following MPP + ‐induced neurotoxicity. Furthermore, HCE effectively modulated key Parkinsonian markers by restoring tyrosine hydroxylase (TH) and reducing the number of α‐syn‐positive cells. At the same concentrations, HCE also attenuated NF‐κB signalling pathway activation and diminished the release of pro‐inflammatory cytokines IL‐1β, IL‐17, and TNF‐α. Notably, HCE promoted the activation of the nuclear receptor peroxisome proliferator activated receptor gamma (PPARγ), known for its neuroprotective properties, and reduced both oxidative and nitrosative stress. Crucially, silencing of PPARγ abolished the beneficial effects of HCE, indicating that its neuroprotective actions are mediated specifically through PPARγ activation. Thus, these findings suggest that HCE confers neuroprotection in vitro by regulating inflammation and oxidative stress primarily via PPARγ modulation. - Aberrant Cell Cycle Gene Expression in a Transgenic Mouse Model of Alzheimer’s Disease

Alzheimer’s disease (AD) is increasingly recognized as a disorder that extends beyond amyloid-β (Aβ) and tau pathology. To this end, growing evidence suggests that aberrant neuronal cell cycle re-entry (CCR) may contribute to neurodegeneration. To investigate this mechanism, we profiled the expression of 84 cell cycle-related genes in the brains of aged APP/PS1 mice, a widely used transgenic model of AD, and compared them with age-matched non-transgenic littermates. Our analysis revealed 32 differentially expressed genes (DEGs), 8 of which exhibited significant changes (fold change > 2, p < 0.05). Several of these DEGs, including CDC7 and CCNC, displayed consistent dysregulation in human AD brains as assessed using the AMP-AD knowledge portal, supporting their translational relevance. Furthermore, integration with miRNA prediction analyses identified candidate post-transcriptional regulators of these DEGs, highlighting novel layers of regulation. Collectively, our results provide the first systematic overview of cell cycle gene dysregulation in aged APP/PS1 mice, establish cross-species concordance with human AD, and propose miRNA–gene interactions as potential contributors to neuronal vulnerability. These findings underscore the importance of cell cycle pathways in AD pathogenesis and point to new avenues for therapeutic exploration. - Neurotransmitter-Mediated Signaling in Glioblastoma and Glial Tumors: Biology and Therapeutic Opportunities

Glioblastoma (GB) is the most common primary malignant brain tumor of adulthood, and despite optimal safe resection and chemoradiation, it is still lethal. Neuroscience of cancer has shown that neuronal activities, as well as neurotransmitters, play an active role in the glioma microenvironment. This article aims to integrate the existing literature on the role of neurotransmitters and their receptors in glioblastoma, as well as other gliomas, highlighting areas of therapeutic intervention in the neuron-tumor interface. We will describe the neuro–glioma interface, including functional neuron–glioma synapses and activity-dependent tumor growth. We will also discuss major neurotransmitter systems involved in glioma pathobiology: glutamate, gamma aminobutyric acid, acetylcholine, dopamine, serotonin, norepinephrine, and other neurotransmitters. We will highlight that these neurotransmitter systems activate common intracellular signaling pathways that control tumor proliferation, invasion, metabolic reprogramming, immune suppression, therapy resistance, etc. In addition, some reports have found tumor-suppressing effects depending on the context. The involvement of neurotransmitter-driven signaling pathways represents a promising area of clinical potential in glioma pathobiology. In particular, focusing on key neurotransmitter systems with blood–brain barrier-permeable agents like alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA/Xc−) system, Muscarinic acetylcholine receptor M3 (CHRM3), dopamine receptor D2, monoamine oxidase A, etc., may enhance drug-repurposing research as well as development of novel anti–neuron–glioma agents. - Troxerutin Potentiated Temozolomide Induced Antitumor Effect in 2D and 3D Glioblastoma Models

Glioblastoma (GBM) is a highly aggressive brain tumour characterised by rapid proliferation and high invasiveness. Temozolomide (TMZ) is the first‐line chemotherapy, but its efficacy is often compromised by intrinsic or acquired resistance and significant side effects. Recent studies have demonstrated that combining TMZ with natural compounds can enhance therapeutic efficacy through synergistic mechanisms and reduce systemic toxicity. In this study, the adjuvant potential of troxerutin (TROX), a natural flavonoid present found in tea, coffee, cereals, fruits and vegetables, and known for its antioxidant and anti‐inflammatory properties, was evaluated in an in vitro model of human U87 cells. The TROX–TMZ combination showed a significant synergistic effect, effectively reducing cell viability, clonogenic capacity, migration and epithelial‐mesenchymal transition (EMT), as well as promoting apoptosis. The combined treatment also modulated oxidative stress and inflammation by activating the KEAP1/NRF2 axis, resulting in increased levels of HO‐1, GSH and SOD1, and decreased ROMO1, ROS, nitrites and pro‐inflammatory cytokines (IL‐6, TNF‐α and IL‐1β). These effects were also confirmed in three‐dimensional models (spheroids), suggesting that the combination exerts significant antitumor activity. These results indicate that TROX could represent an effective adjuvant in the treatment of GBM, providing a novel therapeutic strategy to potentiate the antitumor effects of TMZ. - GIT 27 modulates TLR4/Src/NOX2 signaling pathway: A potential therapeutic strategy to decrease neuroinflammation, oxidative stress and neuronal cell death in Parkinson's disease

BackgroundAphthous stomatitis (AS) is a common idiopathic condition that affects approximately 5% to 25% of the general population. AS is characterized by recurrent painful aphthous ulcers on non‐keratinized oral mucous membranes. Lifestyle modifications and topical treatments are often used to manage symptoms, but no definitive cure exists. This study aims to evaluate the ability of a topical gel containing xyloglucan and pea protein (TXP), two mucoprotective substances, to restore the integrity of oral epithelial cells and manage inflammation associated with AS.MethodsIn this study, oral ulcers were induced in rats by applying a phenol solution for 60 s to the mucosa of the left cheek. Following the formation of the lesion (24 h later) topical treatment with TXP was carried out for 2, 4, and 5 days. The healing properties of TXP were assessed by histopathological examination of the oral mucosa through the evaluation of morphological criteria such as hyperemia, edema, and presence of neutrophilic infiltration and by measuring the expression levels of inflammatory markers such as TNF‐α and IL‐2.ResultsResults demonstrated that TXP was able to effectively treat oral ulcers by reducing the degree of hyperemia and edema in cheek tissue already at day 4. Moreover, TXP significantly decreased the levels of IL‐2 and TNF‐α after 4 days of gel application.ConclusionsThis study demonstrates that TXP possesses a significant ulcer repairing ability in vivo, as it effectively restores the integrity of oral mucosal tissue, thus helping to reduce inflammation. - PF-04691502, a PI3K/mTOR Dual Inhibitor, Ameliorates AD-like Pathology in a Mouse Model of AD

Alzheimer’s disease (AD) is a neurodegenerative disorder that significantly impacts the lives of patients and their families. The pathological features of AD include the accumulation of amyloid-β (Aβ) and Tau, which disrupt neuronal function and communication, ultimately leading to neuronal loss and brain atrophy. Efforts to understand the molecular mechanisms underlying these pathological changes have led to advancements in diagnostic techniques and potential therapeutic interventions. However, the complexity of AD necessitates further research to develop more effective treatments and, ideally, preventive measures. Extensive research suggests that diminishing mTOR signaling increases lifespan and health span across various species. Increased PI3K/mTOR signaling has been linked to the progression of AD pathology, leading to neuronal degeneration and impairments in cognitive function. In this study, we explored the therapeutic potential of PF-04691502, a dual PI3K/mTOR inhibitor, in Alzheimer’s disease (AD)-like pathology using male and female B6.Cg-Tg(APPswe, PSEN1dE9)85Dbo/Mmjax mice (APP/PS1), a well-established transgenic model of AD. Eighteen-month-old APP/PS1 and wild-type mice received oral administration of PF-04691502 at a dose of 1 mg/kg for 12 weeks. Following the treatment period, spatial learning and memory were evaluated using the Morris water maze. Subsequently, the mice brains were collected for neuropathological and biochemical assessments. Our findings showed that PF-04691502 enhanced cognitive performance in APP/PS1 mice and significantly reduced insoluble Aβ accumulation in the brain. Mechanistically, these effects were associated with enhanced autophagy induction. Treatment with PF-04691502 increased the LC3-II/LC3-I ratio, upregulated Beclin-1, and elevated LAMP-2 levels, indicative of stimulated autophagosome formation and lysosomal activity. Overall, these preclinical results suggest that PF-04691502 holds promise as a potential therapeutic agent for AD and other aging-related neurodegenerative diseases involving mTOR pathway dysregulation. - The Role of S6K1 in Aging and Alzheimer’s Disease: Mechanistic Insights and Therapeutic Potential
