He has obtained his graduation degree from Savitribai Phule Pune University and master’s degree (MS Pharm) in Pharmaceutics from National Institute of Pharmaceutical Education and Research-Ahmedabad, an institute of national importance. He has completed his Ph.D. in pharmaceutical Sciences (Pharmaceutics) from Savitribai Phule Pune University. He was awarded with reputed NFST/RGNF Ph.D. JRF/SRF Fellowship for the completion of Ph.D.

Eco-friendly HPTLC method for Trifluridine and Tipiracil determination: Quality-by-design meets green analytical chemistry Amruta Balekundri, Eknath D. Ahire, Rakesh U. Shelke, Dinesh D. Kshirsagar Green Analytical Chemistry, 2025 • An eco-friendly, green chemistry approach was employed using the AGREE scale calculator to ensure sustainability. • The Quality by Design (QbD) methodology, utilizing Analytical Quality by Design (AQbD) tools, was applied to develop a high-performance thin-layer chromatography (HPTLC) method with robust quality attributes. • The method was optimized using Central Composite Design (CCD) under Response Surface Methodology (RSM) for chromatographic parameters. • The validation of the developed HPTLC method was conducted as per ICH Q2 (R1) guidelines, confirming excellent linearity, low limits of detection (LOD) and quantification (LOQ), and high precision (%RSD < 2%). • The environmental impact of the method was evaluated using the ComplexGAPI, AGREE, Eco-Scale, and BAGI tools, yielding high green metrics and confirming minimal environmental impact. Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide, necessitating efficient treatment strategies and accurate analytical methods. This study presents a green analytical approach using a Quality by Design (QbD)-assisted high-performance thin-layer chromatography (HPTLC) method for the simultaneous quantification of trifluridine (TRI) and tipiracil (TIP) in pharmaceutical formulations. Chromatographic parameters were optimized using a Central Composite Design (CCD) under Response Surface Methodology (RSM), with solvent volume and chamber saturation time identified as critical factors. The optimized method yielded reliable Rf values of 0.64 for TIP and 0.91 for TRI. Validation per ICH Q2 (R1) guidelines confirmed excellent linearity (R² = 0.9944 for TIP and R² = 0.9988 for TRI), low detection limits (0.0011 µg/mL for TIP and 0.0022 µg/mL for TRI), and high precision (intra-day %RSD <0.74, inter-day %RSD <0.92). Robustness testing demonstrated minimal variability in Rf values (%RSD <0.28). Environmental sustainability was assessed using ComplexGAPI, AGREE, and BAGI tools. The developed method achieved an AGREE score of 0.81, an Eco-Scale score of 86, and a BAGI score of 80, highlighting its eco-friendliness and practical applicability. This study demonstrates an efficient, precise, and environmentally sustainable analytical method for TRI and TIP quantification, aligning with green chemistry principles and ensuring minimal environmental impact.

Prajapati Applied Nano, 2025 Protein-bound nano-injectable solutions represent a cutting-edge advancement in nanomedicine, offering a versatile platform for precise and controlled drug delivery. By leveraging the biocompatibility and functional versatility of proteins such as albumin, gelatin, and casein, these nano systems enhance drug solubility, prolong circulation time, and improve site-specific targeting, which are particularly beneficial in cancer and inflammatory diseases. This review provides a comprehensive overview of their formulation strategies, physicochemical characteristics, and biological behavior. Emphasis is placed on therapeutic applications, regulatory considerations, fabrication techniques, and the underlying mechanisms of drug–protein interactions. This review also highlights improved pharmacokinetics and reduced systemic toxicity, while also critically addressing challenges like immunogenicity, protein instability, and production scalability. Recent FDA-approved formulations and emerging innovations in precision medicine and theranostics underscore the transformative potential of protein-based nanosuspensions in next-generation drug delivery systems.
Irinotecan's molecular mechanisms against cancer: a primary system biology and chemoinformatics approach for novel formulation development Eknath D. Ahire, Sanjay J. Kshirsagar Current Issues in Pharmacy and Medical Sciences, 2025 Cancer is the third most common type of cancer generally. It affects 6.1% of the entire world’s population and kills 9.2% of all people of both sexes. Even though people with colon cancer have a number of chemotherapies and surgeries to choose from, the disease often returns after the first treatment. AutoDockVina by PyRx 0.8v was used to do molecular docking. The admetSAR2.0 web server was employed for ADMET analysis. The MolSoft and ADVERPred tools were applied to predict the drug's potential for abuse and its potential for side effects. The anti-tumor effects of irinotecan may be aimed at the metabolic processes and Ras and PI3K-Akt signaling pathways that help cancer grow. Gene set enrichment and network analysis proved useful in determining possible protein targets of Irinotecan. Molecular docking revealed how Irinotecan and Vitamin E TPGS interact with EGFR. Moreover, we found that vitamin E TPGS possesses the potential to be an efficient inhibitor for the efflux pump substrate medications such as irinotecan. In addition, the network we created was able to demonstrate how pathways contribute to the protein molecules of irinotecan being able to target cancers. Lastly, we conclude that irinotecan's effectiveness in combating colon cancer is due to the formation of a network of protein-pathway links.
Protective Effect of β-carotene and Turmeric Extract Against Alloxan Generated Oxidative Stress Induced Diabetes Eknath Ahire, Rahul Sable, Rajendra Mogal, Rakesh Shelke, Anjali Tajanpure Biomedical and Pharmacology Journal, 2025 Background and Objective: The present examination was intended to assess the impact of β-carotene and turmeric extract individually, exclusively and with co-administration on antioxidant status in Allxoan instigated diabetic rats. Method: After the administration of β-carotene (20 mg/kg) and turmeric extract (40 mg/kg) exclusively and with co administration, the rodent were contemplated for blood glucose level, liver isolate for cell reinforcement compounds like Superoxide dismutase, Catalase, Lipid peroxidation, diminished glutathione and uric corrosive levels and pancreas for histopathology study. Result: All diabetic groups show significantly elevated blood glucose levels compared to the normal control, confirming hyperglycemia. Blood glucose levels increase further (300.56 ± 12.64 mg/dL), indicating worsening hyperglycemia. The combination shows the most substantial reduction in glucose levels (175.25 ± 3.80 mg/dL), nearing normal values, highlighting a synergistic effect. The combination significantly reduces LPO (36.01 ± 0.837), nearing normal levels, suggesting a powerful reduction in oxidative damage. The combination shows the highest improvement among treatments in SOD (58.97 ± 1.26), CAT (0.931 ± 0.025), GSH (32.66 ± 0.554), suggesting a synergistic effect in enhancing enzymatic antioxidant defense. The combination of β-carotene and turmeric extract (7.612 ± 0.121 g) nearly normalizes liver weight, suggesting a synergistic effect in protecting the liver from diabetes-induced damage. The simultaneous administration of these drugs significantly rejuvenates pancreatic beta cells. Conclusion: The outcomes got from this investigation gives a logical confirmation that β-carotene and turmeric separate have defensive impact against alloxan creates oxidative pressure prompted diabetes. The combination treatment shows synergistic effect.
