  • Study of Local Black Iraqi Goats Genotypes for the Cytb Gene

    Goats are the earliest domesticated ruminants. The local goat, Capra hircus, is considered one of the most important animals globally to provide good livestock production under harsh environmental conditions. This study aimed to detect the genetic structures of the local Iraqi goats bred in the central and southern regions of the country and investigate the possibility of benefiting from their genetic structures to construct improvement programs for increasing the productivity of these animals. To this end, blood samples were taken from 15 domestic black goats. A total of 10 ml of each animal's blood was placed in plastic containers of 10 ml. The DNA was extracted and sent to the laboratories of Juan Ju University, People's Republic of China, to analyze the sequences of the nitrogenous bases of the Cytochrome b (Cytb) gene. The results showed the presence of a genetic morphology for a segment of 670 base pairs for all the studied samples, and 15 sequences of this strain were recorded in the gene bank under the following accession numbers (LC496353.1:1-LC496367.1:1). The sequences of the nitrogenous bases of this segment of the gene, which were registered in the gene bank of some international goat breeds, were used for comparison with the sequences of black Iraqi goats to analyze the phylogenetic tree, calculate the genetic distance, study haplotypes, and calculate neutrality. The results showed the presence of one mutation in the studied segment of the Cytb gene, with a size of 670 bp. The mutation in base 46 of the studied gene converted from the purine group to the pyrimidine group (the shift from the nitrogen leaders A<C) in all the studied samples. It led to the transformation of the amino acid Asparagine into Histidine, where 233 amino acids were obtained, dominated by the amino acid Isoleucine and Leucine, over other amino acids at a rate of 14.34% and 11.21%, respectively. The phylogenetic tree showed the existence of two main branches, one of which included the local black Iraqi goat breed, and the other included all the international breeds under comparison. It is concluded that the black Iraqi goat breed has a different origin from other breeds.
  • Relationship between various meat qualities, tenderness, and Jenoubi and crossbreed cows' CAPN1 gene polymorphism

    139 calves of the Jenoubi breed and crossbred were used (73 and 66 calves, respectively), their ages ranging between 2-3 years, from Thi- Qar governorate. Blood and meat samples were collected, and tests were performed on them. The polymerase chain reaction (PCR) and sequencing techniques were used to detect the gene genotypes. The results showed moderate to high allelic frequencies that correlate with the tenderness of the CAPN1 gene. The two genotypes, GG and AG, showed significant variations (P&lt; 0.05) in most sensory traits at sites 316 and 530, including tenderness, juiciness, and general acceptability. Additionally, the genotype GG within the Jenoubi breed significantly outperformed in pH, water holding capacity, and cooking loss, with values of 5.81, 7.65, and 49.76, respectively. While the GG genotype of cross animals at site 316 showed better water-holding capacity and cooking loss. There are relationships between genotypes of this gene in two different sites so that it might be used in the selection program of local and their crosses.   • Using of Microsatellites to Study the Genetic Polymorphisms of SRYm18 Region in Iraqi Sheep

    The origin of animals is usually determined by their paternal genotypes of the genes on the Y chromosome. In addition to the genes and their polymorphisms in the genome of mitochondria that are inherited through dams. In view of the lack of studies focusing on the genes of the Y chromosome in the world and their absence in Iraq. The aim of the present study was to identify the multiple genetic polymorphisms of the SRYM18 gene in the Arabi and Awassi sheep raised in Iraq. The study was conducted in the Genetic Engineering Laboratories - College of Agriculture, University of Basra, as well as in the Basra Genome Laboratory. The amplification of the SRYM18 gene showed genetic polymorphisms and gave a gene segment of (103-880) bp. The number of alleles of the SRYm18 gene was 13 alleles in the Arabi and 16 alleles in the Awassi breeds. The equilibrium test showed that the two breeds were under equilibrium. The two breeds were identical with nine alleles, while the number of special alleles for the Arabi breed was two, while the Awassi breed was distinguished by five. The number of rare alleles reached 20, of which seven were of the Arabi breed, and 13 of them were of the Awassi breed. Mean expected heterozygosity was 0.6386 with nonsignificant Fis for Arabi breed (0.1541) but significant for Awassi breed (0.2213). Mean neutrality was close to lower bound (0.1721) and (0.1270) for Arabi and Awassi breeds respectively

    The present study was undertaken to characterize the genetic diversity of the aromatase cytochrome P450 (CYP19) gene in 34 cows (15 local, 14 Holstein, and 5 Crosses) in Iraq. The objectives of the present study are to detect SNPs (mutations) in promoter p1.1 of the CYP19 gene in cattle bred in Iraq using sequencing techniques. We identified five single-nucleotide polymorphisms (SNP) loci of the CYP19 gene that were detected, namely G933T, G994C, A1044G, A1062T, and C1468A. The results showed the presence of 3, 4, and 2 polymorphic sites leading to the construction of 4, 5, and 3 different haplotypes for Holstein, local, and crosses respectively. Haplotype diversity were 0.791, 0.752, and 0.700 respectively. While nucleotide diversity was 0.0017, 0.0022, and 0.0013 respectively. Besides, we carried out a phylogenetic analysis of these sequences to address the evolutionary relationship between the animal species. These fragments were assigned in the GenBank database under the accession numbers: LC490756, LC490757, LC491437, LC491438, LC491439, LC491588, and LC491589.

  • Impact of the Iraq marshlands restoration program on livestock population and production in the southern marshes of Iraq

    The USAID Iraq Marshlands Restoration Program (IMRP) promoted the application of two promising initiatives tested from 2004 to 2006: (1) the introduction of alfalfa and maize fodder to feed and improve the nutrition of buffalo and (2) the creation of a veterinary service to increase livestock production and health. The livestock sector in the marshlands has seen dramatic improvement since IMRP ended in 2006. These changes are across the board in the absolute number of livestock (from 66,700 heads in 2006 to 111,525 heads in 2017) and in their increasing diversity (buffalo were 74.33% of all livestock in 2006 but decreased to 70.33% in 2017). Buffalo, specifically, had greater birth weight (40 kg in 2006 and 45.6 kg in 2017), greater daily growth rates (0.3 kg in 2006 and 1-1.25 kg in 2017), higher daily milk yields (8.81 kg/day in 2006 and 9.69 kg/day in 2017), and longer lactation periods (150 days in 2006 and 238 days in 2017). There have been accompanying advances in the closely related meat and dairy markets. Live weight and market prices for buffalo and cattle have increased three- to four-fold. Rising daily milk yields also sparked the creation of cottage industries to produce dairy products using higher fat buffalo milk. These changes all point to an increasingly healthy and dynamic livestock sector in the marshlands. However, other anticipated changes in the marshlands did not meet expectations. In 2006, IMRP predicted a strong shift away from the historical dominance of buffalo ownership to a more diversified pattern of ownership that included the relatively newly introduced cattle and a rapid increase in sheep holdings, making the latter the dominate animal. Instead, buffaloes have continued to be the vastly most numerous animals owned in the marshlands. In 2017, they ranged from 65 to 75% of all livestock holdings. Greater access to reflooded marshes by buffalo herders and increased financial earnings realized from meat and dairy production may help to explain the continued reliance on buffalo ownership. IMRP’s introduction of a veterinary service dedicated to working in the marshlands villages was far less successful but highly regarded by Marsh Arabs, being the first time that the services were offered in the marshes.
